Striatal GABAergic interneurons (GINs) provide rapid corticostriatal feedforward inhibition to SPNs, while cholinergic interneurons (CINs) inhibit SPNs by activating nicotinic acetylcholine receptors (nAChRs) on GINs. The disynaptic inhibitory pathway mediated by nAChRs is slower than direct GIN-mediated feedforward inhibition, and the role of nAChRs in striatal synaptic integration is unclear.

Differentiating the Neuropharmacological Properties of Nicotinic Acetylcholine Receptor-Activating Alkaloids

Summary: This study examined the neuropharmacological effects of natural alkaloids, including nicotine, cotinine, and anatabine. Nicotine showed anxiolytic-like behavioral effects in mice and zebrafish, while cotinine and anatabine had no detectable effects. Nicotine stimulated the release of norepinephrine and dopamine, while cotinine and anatabine mainly stimulated dopamine release. These findings suggest that natural nicotine alkaloids can induce an anxiolytic-like behavior in nonclinical animal models, which may depend on the activation of various nicotinic acetylcholine receptors and regulation of neurotransmitter systems.

Summary: Mutant subunits of the neuronal nicotinic ACh receptor can cause Autosomal Dominant Sleep-related Hypermotor Epilepsy (ADSHE), characterized by frontal seizures during NREM sleep. The study found that ADSHE may be related to prefrontal hyperexcitability and glutamatergic deficit. Mutant mice showed a decrease in excitatory input to FS interneurons, but an increase in inhibition by somatostatin-expressing cells. These findings explain why NREM sleep favors ADSHE seizures and suggest that nicotine may be palliative in patients.

Genetic Variant in Nicotinic Receptor α4-Subunit Causes Sleep-Related Hyperkinetic Epilepsy via Increased Channel Opening
