Polymer- and lipid-based nanocarriers for ocular drug delivery: Current status and future perspectives

Haijie Han et al.

Summary: Ocular diseases have a significant impact on patients' vision and quality of life, with a global prevalence of over 43 million blindness. However, delivering drugs effectively to the eyes, especially for intraocular disorders, remains a major challenge due to various ocular barriers. Recent advancements in nanocarrier technology provide a promising solution by improving drug penetration, retention, solubility, toxicity reduction, release duration, and targeted drug delivery. This review highlights the progress and applications of nanocarriers, specifically polymer- and lipid-based nanocarriers, in treating different eye diseases and their potential for efficient ocular drug delivery. It also discusses ocular barriers, administration routes, and future prospects and challenges in the field of nanocarriers for ocular disease treatment.

Poloxamer 188-quercetin formulations amplify in vitro ganciclovir antiviral activity against cytomegalovirus

Andrew Kjar et al.

Summary: The study investigated the use of poloxamer 188 and quercetin as bioactive excipients to reduce the dosage of ganciclovir (GCV) in the treatment of human cytomegalovirus (CMV) infection. The combination of GCV with the poloxamer-solubilized quercetin showed modest CMV viral inhibition, but when coadministered with GCV, it exhibited an additive or synergistic interaction in subtherapeutic ranges of GCV. This enhanced GCV antiviral therapy may provide a viable approach to reduce GCV dosage and associated toxicity.

Topical ocular delivery of vancomycin loaded cationic lipid nanocarriers as a promising and non-invasive alternative approach to intravitreal injection for enhanced bacterial endophthalmitis management

Kamyar Jounaki et al.

Summary: NLCs fabricated VCM carriers can effectively enhance the ocular penetration and residence time of vancomycin, showing significant sustained drug release. The optimized formula significantly increased intravitreal concentration of VCM after ocular administration, showing comparable therapeutic effects to intravitreal injection, with nonirritating and safe profile.

To investigate fit-to-purpose nanocarrier for non-invasive drug delivery to posterior segment of eye

Vishakha Tambe et al.

Summary: Diseases of the posterior segment of the eye are challenging to treat due to limited drug bioavailability. This study aimed to explore a non-invasive nanocarrier system for efficient drug delivery. Results showed that DEX-NLCs had the highest permeation percentage through the cornea, while DEX-CUBs exhibited no cytotoxicity in ARPE-19 cells. NLCs and NMFs were selected as the most promising carriers for further studies based on their superior permeation potential in both in vivo and in vitro studies.
