Diagnosis and follow-up of glycogen storage disease (GSD) type VI from the largest GSD center in China

Xiaomei Luo et al.

Summary: This study investigated the clinical profiles, molecular diagnosis, and treatment outcomes of patients with GSD VI. The main initial clinical features of this disease include hepatomegaly, short stature, elevated liver transaminases, etc. Uncooked cornstarch treatment significantly improved the stature and biochemical parameters. The study also identified novel variants of PYGL and found two hot-spot variants in China.

Hepatic glycogen storage diseases type 0, VI and IX: description of an italian cohort

Francesco Tagliaferri et al.

Summary: A study on 30 Italian patients with GSD revealed that prompt establishment of specific nutritional therapy is crucial in preserving growth, improving glycemic control, and preventing liver complications. High protein diet may have a protective effect against overweight/obesity and insulin resistance.

Liver histology in children with glycogen storage disorders type VI and IX

Irene Degrassi et al.

Summary: This study retrospectively reviewed 22 children diagnosed with GSD VI and IX at a pediatric liver center between 2001 and 2018. Despite GSD VI and IX being considered clinically mild, varying degrees of chronic histological changes were observed in all liver biopsies. Histopathological assessment of liver involvement was found to be superior to biochemical parameters for definitive classification, though mutational analysis is still necessary.

Whole-Exome Sequencing Uncovers Novel Causative Variants and Additional Findings in Three Patients Affected by Glycogen Storage Disease Type VI and Fanconi-Bickel Syndrome

Maryam Eghbali et al.

Summary: This study evaluated the use of whole-exome sequencing for genetic analysis of liver GSD-suspected patients and discovered three novel causative gene variants, enhancing diagnostic accuracy for GSD.

Identification and Characterization of a Novel Splice Site Mutation Associated with Glycogen Storage Disease Type VI in Two Unrelated Turkish Families

Sarah C. Grunert et al.

Summary: The study reported novel homozygous splice site variants in two Turkish patients with GSD VI, leading to impaired protein stability. In the era of next generation sequencing, techniques like transcriptome analysis are crucial for confirming pathogenicity and determining personalized therapeutic measures based on genetic results.

The Phenotypic and Genetic Spectrum of Glycogen Storage Disease Type VI

Sarah Catharina Gruenert et al.

Summary: Glycogen storage disease type VI (GSD VI) is a disorder with broad clinical heterogeneity, a small number of patients with severe phenotype and liver cirrhosis, and neither clinical nor laboratory findings allow for a differentiation between GSD VI and GSD IX. Early biochemical markers of disease severity or clear genotype phenotype correlations are missing.
