Synthesis of Indenones Via Palladium-Catalyzed Carbonylation with Mo(CO)6 as a CO Surrogate

Summary: This paper reports an effective ligand-free Pd-catalyzed carbonylation of o-bromoaryl iodides with alkynes to obtain indenone compounds. The reaction shows good tolerance towards functional groups on o-bromoaryl iodides and alkynes. By introducing Mo(CO)6 as a carbonyl source, the reaction becomes safer and more operable. Mechanistic investigations reveal a sequential process of oxidative addition, alkyne insertion, carbonyl insertion, and reductive elimination to produce the desired carbonylation products.

The use of cheap and easy-to-handle Na2S·9H2O as the reductant in the presence of PPh3 and N-chlorosuccinimide (NCS) enables the selective dehydroxylation of alpha-hydroxyl carbonyl compounds, including ketones, esters, amides, imides, and nitrile groups. The synthetic utility is demonstrated by the late-stage deoxygenation of bioactive molecule and complex natural products.

A variety of alkyl and halogen substituted amides were prepared in moderate to good yields.

In this study, seven DMSO complexes were screened and their structures confirmed, with some showing unique arrangement diversities compared to known DMSO complexes and parent motifs. Experimental data suggested that planarization of aryl planes in diarylureas may play a crucial role in cocrystallization.
