Rh(III)-Catalyzed C-H Functionalization/Annulation of 1-Arylindazolones: Divergent Synthesis of Fused Indazolones and Allyl Indazolones

Bing Shu et al.

Summary: Rh(III)-catalyzed C-H/N-H annulation and C-H allylation of phenylindazolones have been achieved using 5-methylene-1,3-dioxan-2-one and 4-vinyl-1,3-dioxolan-2-one as scalable cross-coupling partners, providing functionalized indazolone fused heterocycles and branched and linear allyl indazolones with moderate to high yields. These synthetic protocols demonstrate mild conditions, broad substrate scope, and high functional-group compatibility. Additionally, scale-up synthesis and preliminary mechanistic exploration were also conducted.

Copper(I)-Mediated Cascade Annulation via Dual C-H/C-H Activation: Access to Benzo[a]carbazolic AEEgens

Tamanna Khandelia et al.

Summary: A Cu(I)-mediated cascade cyclization/annulation of unprotected o-alkynylanilines with maleimides in one pot is developed, providing fused benzo[a]carbazoles with free NH skeletons. The annulated products exhibit fluorescence emission in the range of 485-502 nm, with a large Stokes shift and fluorescence lifetime of around 17 ns. Cell viability assays confirm the biocompatibility of the annulated products and their potential for cell imaging applications.

Access to Branched Allylarenes via Rhodium(III)-Catalyzed C-H Allylation of (Hetero)arenes with 2-Methylidenetrimethylene Carbonate

Shang-Shi Zhang et al.

Summary: A novel rhodium(III)-catalyzed C-H allylation reaction has been developed using 2-methylidenetrimethylene carbonate as an efficient allylic source, resulting in the synthesis of various branched allylarenes bearing an allylic hydroxyl group in moderate to excellent yields. Five different directing groups, including oxime, N-nitroso, purine, pyridine, and pyrimidine, were found to be compatible in this reaction.

Rh(III)-Catalyzed [5+1] Annulation of Indole-enaminones with Diazo Compounds To Form Highly Functionalized Carbazoles

Zhidong Jiang et al.

Summary: A novel Rh(III)-catalyzed C-H activation/annulation cascade of indole-enaminones with diazo compounds was reported, generating diversely functionalized carbazole frameworks with good to excellent yields. The most notable feature is the formation of a novel [5 + 1] cyclization product, accompanied by the complete removal of acetyl and N,N-dimethyl groups from the substrates, rather than the expected [4 + 2] cyclization products.

Yuanfei Zhang et al.

Summary: The Rh(III)-catalyzed synthesis method has been achieved for preparing isoquinolinones containing a C3 quaternary center, which is applicable to late-stage diversification of N-methoxyamides derived from steroids and adapalene. This approach serves as a key step for conveniently synthesizing US28 inverse agonist analogs.

Rh(iii)-Catalyzed tandem C(sp2)-H allylation/N-alkylation annulation of arene amides with 2-alkylidenetrimethylene carbonates

Hui Xie et al.

Summary: A Rh(iii)-catalyzed tandem reaction using 5-methylene-1,3-dioxan-2-one as an efficient allyl source has been developed for the synthesis of 3,4-dihydropyrimido[1,6-a]indol-1(2H)-ones, showing good substrate scope and moderate to excellent yields. The Rh(iii) catalyst plays a dual role in C-H activation and N-alkylation annulation process in the mechanism studies.

Direct Synthesis of Benzo[c]carbazoles by Pd-Catalyzed C-H [4+2] Annulation of 3-Arylindoles with External 1,3-Dienes

Baptiste Gerardin et al.

Summary: In this study, a regioselective Pd-catalyzed C-H [4 + 2] benzannulation of N-unprotected 3-arylindoles with external readily available 1,3-dienes was presented. Depending on the nature of the solvent, DCE or CH3CN, two consecutive approaches, oxidative or reductive, were validated to design a novel library of C6 alkyl or (E)-C6-styryl-benzo[c]carbazoles in moderate to good yields.
