Fluorine-containing drugs approved by the FDA in 2021

Summary: This review article presents nine newly approved fluorine-containing drugs by the US Food and Drug Administration (FDA) in 2021. These small molecule drugs feature aromatic fluo-rine, trifluoromethyl, and chlorodifluoro groups. The therapeutic areas of these fluorine-containing drugs include multiple myeloma, lymphoma, HIV, chronic heart failure, chronic myeloid leukemia, (ANCA)-associated vasculitis, migraines, von Hippel-Lindau disease, and non-small cell lung cancer. The review also discusses the brief biological activities and synthetic methods for each of these nine drugs.

Nickel-Catalyzed anti-Markovnikov Hydrodifluoroalkylation of Unactivated Alkenes

Summary: An efficient Ni-catalyzed hydrodifluoroalkylation of unactivated alkenes with bromodifluoroacetate by using PhSiH3 as hydride source has been developed. The transformation provides aliphatic difluorides with anti-Markovnikov regioselectivity. A wide range of substrates, including highly remote alkenes, drug molecules, commercially available CF2 precursors, and even nonfluorinated substrates, are competent in this reaction under mild conditions, demonstrating the practicality of the strategy. Mechanistic studies suggest that the difluoroalkyl radical may be a key intermediate in this transformation.

Photocatalytic Reductive Olefin Hydrodifluoroalkylation Enabled by Tertiary Amine Reductants Compatible with Complex Systems

Catalytic Hydrodifluoroalkylation of Unactivated Olefins

Remote Nucleophilic Substitution at a C(sp3)-H Bond of a-Bromocarboxamides via 1,4-Hydrogen Atom Transfer To Access N-Acyl-N,O-acetal Compounds

Summary: Here, we report a method for synthesizing N-acyl-N,O-acetal moieties via a remote nucleophilic substitution reaction using 1,4-hydrogen atom transfer (1,4-HAT). The reaction proceeds through rS(N) at a C(sp(3))-H bond of α-bromocarboxamide in the presence of an alcohol/phenol and a copper catalyst. Mechanistic studies confirmed that the 1,4-HAT process is the rate-determining step. The rS(N) reaction can be employed with different α-bromocarboxamides for the selective synthesis of topologically complex N-acyl-N,O-acetal molecules.

Visible-Light-Induced Metal-Free Decarboxylative Perfluoroalkylation of Aryl Acrylic Acids

The easy accessibility of perfluoroalkyl iodides and the broad substrate scope, mild conditions, and metal-free catalyst make this protocol applicable for transforming inexpensive raw materials to high-value chemicals.

Palladium-catalyzed difluoroalkylative carbonylation of styrenes toward difluoropentanedioates

Summary: The introduction of fluorine atoms into organic molecules is an attractive topic. This work developed an interesting palladium-catalyzed reaction to transform aryl olefins into difluoropentanedioate compounds with good functional-group tolerance and high regioselectivity. Ethyl bromodifluoroacetate acts as both a difluoroalkyl precursor and a nucleophile in this reaction. Additionally, successful scale-up reactions and further transformations of the obtained product were demonstrated.
