Metallaphotoredox: The Merger of Photoredox and Transition Metal Catalysis

Amy Y. Chan et al.

Summary: The merger of photoredox catalysis with transition metal catalysis, known as metallaphotoredox catalysis, has become an important part of synthetic methodology. It combines the bond formation capability of transition metal catalysis with the utility of photoinduced electron and energy transfer processes. Photocatalytic substrate activation allows the involvement of simple starting materials in metal-mediated bond-forming processes, while electron or energy transfer with organometallic intermediates provides a complementary activation mode to traditional catalytic platforms.

Photoredox-Catalyzed C-H Functionalization Reactions

Natalie Holmberg-Douglas et al.

Summary: The fields of C-H functionalization and photoredox catalysis have gained significant interest and utility in various scientific disciplines. This review focuses on the use of photoredox catalysis in C-H functionalization reactions, highlighting both inorganic/organometallic and organic-based photoredox catalytic systems. The subdivision by reaction class (sp(2) or sp(3) C-H functionalization) provides perspective and tactical strategies for synthetic applications.

Accelerating reaction generality and mechanistic insight through additive mapping

Cesar N. Prieto Kullmer et al.

Summary: The generality of a reaction is crucial for determining its overall impact and usefulness in synthetic methods. This study developed an additive mapping approach that rapidly expands the utility of synthetic methods and provides mechanistic insight. By validating this approach on a specific reaction, the discovery of a phthalimide ligand additive was made, which overcomes limitations and has important mechanistic implications for nickel-catalyzed cross-couplings.

Tetrahydrofuran-Based Transient Receptor Potential Ankyrin 1 (TRPA1) Antagonists: Ligand-Based Discovery, Activity in a Rodent Asthma Model, and Mechanism-of-Action via Cryogenic Electron Microscopy

Jack A. Terrett et al.

Summary: This study optimized a series of potent TRPA1 small molecule antagonists, leading to the discovery of a novel tetrahydrofuran-based linker. Compound 20 showed significant dose-dependent reduction of inflammatory response in a rat pain model and a guinea pig asthma model, and the structure of the TRPA1 channel bound to compound 21 was determined via cryogenic electron microscopy.
