A Traceless Heterocyclic Amine Mediator in Regioselectivity-Switchable Formal [1+2+2] Cycloaddition Reaction to 1,3,4-and 1,4,5-Trisubstituted Pyrazoles

Summary: Switchable multicomponent reactions have been developed to construct compound libraries with diverse and complex skeletons by altering the reaction conditions. This study presents a regioselectivity-switchable formal [1 + 2 + 2] cycloaddition reaction using difluoroalkyl compounds, enaminones, and RNHNH2, with 1-methylindazol-3-amine as a traceless mediator. The reaction switches the inherent regioselectivity from 1,3,4-trisubstituted pyrazoles to 1,4,5-trisubstituted pyrazoles. Notable features include mild conditions, simple operation, and broad scopes.

Amine-catalyzed synthesis of N2-sulfonyl 1,2,3-triazole in water and the tunable N2-H 1,2,3-triazole synthesis in DMSO via metal-free enamine annulation

Summary: Selective synthesis of N-2-sulfonyl and N-2-H1,2,3-triazoles was achieved through organocatalytic annulation of enaminone/enaminoester with sulfonyl azide. The unconventional selectivity for N-2-sulfonyl 1,2,3-triazoles occurred in pure water, where the hydrogen bond effect between water and the intermediate formed by enamine-azide corporation accounted for the novel reaction selectivity. In contrast, the reactions conducted in DMSO specifically yielded N-2-H 1,2,3-triazoles in the absence of such hydrogen bond effect.

Ultrasound-Promoted Synthesis of α-Thiocyanoketones via Enaminone C=C Bond Cleavage and Tunable One-Pot Access to 4-Aryl-2-aminothiazoles

Palladium-Catalyzed Three-Component Selective Aminoallylation of Diazo Compounds

Summary: In this study, a Xantphos-containing dinuclear palladium complex was used to catalyze the geminal aminoallylation of diazocarbonyl compounds, providing a selective synthesis of various quaternary alpha-amino esters. The reaction did not proceed through direct N-H insertion, allylic alkylation of amino nucleophiles, or diene formation. Mechanistic investigations indicated that a relayed pathway via allylation or a [2,3]-sigmatropic rearrangement was unlikely.

Inverse Electron Demand Diels Alder Reaction of Aza-o-Quinone Methides and Enaminones: Accessing 3-Aroyl Quinolines and Indeno[1,2-b]quinolinones

Summary: We have developed a new synthetic method, employing a Diels Alder cycloaddition reaction to generate 3-aroyl quinolines from enaminones and aza-o-quinone methides. The general applicability of this reaction was demonstrated with various substituted enaminones and aza-o-quinone methides, and the methodology was successfully validated in gram scale. Additionally, we carried out a one-pot strategy to synthesize 3-acyl quinolines from aliphatic ketones. Finally, the 3-aroyl quinolines were used for the synthesis of indeno[1,2-b]quinolinones via a Pd-catalyzed dual C-H activation approach.

Rh(III)-Catalyzed Enaminone-Directed C-H Coupling with Diazodicarbonyls for Skeleton-Divergent Synthesis of Isocoumarins and Naphthalenes

Summary: Diversity-oriented synthesis is a valuable method for expanding the chemical space, but its application is limited by the available toolbox for skeleton-diversification chemistry. In this study, we report the Rh(III)-catalyzed coupling of enaminones and diazodicarbonyls for the synthesis of isocoumarins and naphthalenes with divergent skeletons. The dependence of the skeleton-forming process on the ring size of diazodicarbonyls and pH demonstrates the achievement of both substrate-and reagent-controlled skeletal diversity generation in a single system. Moreover, the C-C bond cleavage reactivity is found to be critical for facile synthetic access to isocoumarins.

The merger of vinyl-N-triftosylhydrazones and silver catalysis to enable stereoselective vinylcyclopropanation of alkenes

Summary: In this study, a strategy combining silver catalysis was developed to address the challenge of competitive pyrazole formation from vinyl-N-sulfonylhydrazones, enabling the selective formation of highly electrophilic silver vinylcarbenoid species. This strategy allowed for a powerful silver-catalyzed stereoselective vinylcyclopropanation of alkenes, with diverse substrates and high yields. The usefulness of this method was demonstrated in gram-scale synthesis, late-stage modification, and total synthesis of bioactive molecules.

An expedient synthesis of highly functionalized 1,3-dienes by employing cyclopropenes as C4 units
