Design and Application of m-Hydroxybenzyl Alcohols in Regioselective (3+3) Cycloadditions of 2-Indolymethanols†

Yi-Cheng Shi et al.

Summary: A new class of m-hydroxybenzyl alcohols has been designed as competent three-carbon building blocks and successfully applied in a series of reactions. These reactions can synthesize indole-fused six-membered rings with good yields and selectivity. Furthermore, a preliminary investigation of the enantioselective version of these reactions has shown the potential for constructing chiral indole-fused six-membered rings.

Design and synthesis of axially chiral aryl-pyrroloindoles via the strategy of organocatalytic asymmetric (2

Ping Wu et al.

Summary: In this study, a new strategy for designing and synthesizing axially chiral indole-based frameworks was developed. Aryl-pyrroloindoles were synthesized via organocatalytic asymmetric cyclization between 3-arylindoles and propargylic alcohols. These frameworks were also transformed into efficient chiral ligands for palladium-catalyzed asymmetric reactions. The reaction mechanism was investigated using theoretical calculations.

Nickel-Catalyzed Stereo- and Enantioselective Cross-Coupling of gem-Difluoroalkenes with Carbon Electrophiles by C-F Bond Activation

Ziqi Zhu et al.

Summary: The stereo- and enantioselective cross-electrophile coupling involving C-F bond activation was successfully achieved using a chiral nickel complex and B(2)pin(2) as a stoichiometric reductant. The reaction allows the construction of a C(sp(2))-C(sp(3)) bond under mild conditions, resulting in a variety of monofluoroalkenes bearing stereogenic allylic centers. Initial mechanistic studies suggest a radical chain pathway may be involved in the C-F bond activation process.

Asymmetric Boration of para-Quinone Methides Catalyzed by N-Heterocyclic Carbene

Wu Yuzhu et al.

Summary: The first carbene-catalyzed enantioselective 1,6-conjugate addition of boron to para-quinone methides was achieved using a commercially available chiral triazolium and bis(pinacolato)diboron. This reaction features mild conditions, broad substrate scope, excellent functional group tolerance, and good yield and enantioselectivity. Remarkably, even in the absence of any transition metals, the reaction proceeded well with just 0.1 mol% of catalyst loading without significant loss of yield and enantioselectivity, although longer reaction time was required.

Stereochemical Control via Chirality Pairing: Stereodivergent Syntheses of Enantioenriched Homoallylic Alcohols

Shang Gao et al.

Summary: This study presents the development of stereodivergent syntheses of enantioenriched homoallylic alcohols using a chiral nonracemic alpha-CH(2)Bpin-substituted crotylboronate. The catalyst-controlled stereoselectivity allowed for the synthesis of complementary stereoisomers of chiral homoallylic alcohols from the same boron reagent. Computational studies were conducted to investigate the origins of the observed stereoselectivity, and the resulting products are valuable for the rapid construction of polyketide structural frameworks.

Difluorocarbene-Induced Ring-Opening Difluoromethylation-Halogenation of Cyclic (Thio)Ethers with TMSCF2X (X=Br, Cl)**

Rongyi Zhang et al.

Summary: A simple strategy using difluorocarbene to activate carbon-chalcogen bonds is reported for the ring-opening difluoromethylation-halogenation of cyclic (thio)ethers. This reaction proceeds through the in situ protonation of previously little-known difluoromethylene oxonium or sulfonium ylide intermediates, followed by ring-opening with halide ion to yield halogenated acyclic difluoromethyl (thio)ethers. TMSCF2X (X=Br, Cl) serves as unique reagents for both difluorocarbene source and halide ion source in this synthetic purpose.
