Copper-Catalyzed Asymmetric Diyne Cyclization via [1,2]-Stevens-Type Rearrangement for the Synthesis of Chiral Chromeno[3,4-c]pyrroles

Summary: In this study, a copper-catalyzed asymmetric cascade cyclization/[1,2]-Stevens-type rearrangement was reported to efficiently assemble a variety of valuable chiral chromeno[3,4-c]pyrroles bearing a quaternary carbon stereocenter with generally moderate to good yields and excellent enantioselectivities (up to 99% ee). Notably, this protocol represents the first example of catalytic asymmetric [1,2]-Stevens-type rearrangement based on alkynes and the first asymmetric formal carbene insertion into the Si-O bond.

[4+1] Cyclization of benzohydrazide and ClCF2COONa towards 1,3,4-oxadiazoles and 1,3,4-oxadiazoles-d5

Summary: A facile synthesis method for 1,3,4-oxadiazoles and 1,3,4-oxadiazoles-d5 via [4 + 1] cyclization of ClCF2COONa with non-amine compounds containing amino groups is developed. This is the first time halofluorinated compounds are used as C1 synthon to construct deuterated nitrogen-heterocyclic compounds. The current protocol features simple operation, readily accessible raw materials, wide substrate scope, and valuable products.

Summary: In this study, a Lewis acid-catalyzed [4 + 2] cycloaddition reaction between 3-alkyl-2-vinylindoles and beta,gamma-unsaturated alpha-ketoesters was successfully established in the presence of Sc(OTf)(3). This method provided a series of indole-containing pyran derivatives with generally good yields (up to 90% yield) and excellent diastereoselectivities (up to > 95:5 dr) under mild conditions. The research not only enriches the chemistry of 3-alkyl-2-vinylindoles, but also provides an atom-economic method for the synthesis of indole-containing pyran derivatives with potential bioactivity.

Biomimetic Carbene Cascades Enabled Imine Derivative Migration from Carbene-Bearing Thiocarbamates

Summary: Inspired by the body circulation of Omeprazole, a novel carbenetriggered cascades for synthesizing 2-aminobenzofuran derivatives from N-sulfonyl-1,2,3-triazoles or benzothioazole-bearing thiocarbamates was disclosed, representing an unprecedented imine derivative migration process. Additionally, desulfurizing reagent-free Barton-Kellogg-type reactions starting from N-sulfonyl-1,2,3-triazoles were achieved for the first time, with elemental sulfur confirmed as a byproduct. Experimental data and DFT calculations further explained the unique reactivity observed in these reactions.
