Enhanced axial resolution of lattice light sheet microscopy by fluorescence differential detection

Summary: Lattice light-sheet microscopy (LLSM) is a promising technique for long-term biological volumetric imaging. However, improving the axial resolution in 3D imaging remains a challenge. Combining LLSM with fluorescence differential detection, known as LLSDM, has been proposed to enhance the axial resolution by discarding off-focus photons. This study modifies the LLSDM theory to generate an ideal complementary lattice pattern with improved symmetry and demonstrates its effectiveness in improving axial resolution and imaging contrast compared to traditional LLSM.

Deep low-excitation fluorescence imaging enhancement
