Comprehensive genetic analysis using next-generation sequencing for the diagnosis of nephronophthisis-related ciliopathies in the Japanese population

Nana Sakakibara et al.

Summary: Nephronophthisis is a common kidney disease related to abnormalities in primary cilia. This study identified multiple causative genes for NPHP-RCs using genetic analysis, and found that most patients showed extrarenal manifestations. Comprehensive genetic analysis using NGS is useful for diagnosing patients with NPHP-RCs.

The Impact of Inherited Retinal Diseases in the United States of America (US) and Canada from a Cost-of-Illness Perspective

Jennifer Gong et al.

Summary: The study estimated the annual costs of inherited retinal diseases (IRDs) in the US and Canada from a societal perspective, including economic costs and wellbeing costs. The findings showed that IRDs impose significant economic and wellbeing costs in both countries, with wellbeing costs accounting for over 60% of the total costs. Further research into cost-effective therapies and interventions for vision loss from IRDs is needed.

LEBER CONGENITAL AMAUROSIS DUE TO CEP290 MUTATIONS-SEVERE VISION IMPAIRMENT WITH A HIGH UNMET MEDICAL NEED A Review

Bart P. Leroy et al.

Summary: LCA10, caused by CEP290 mutations, is a genetic retinal disease leading to severe visual impairment in early childhood. There are currently no approved treatments, but investigational strategies include RNA editing using antisense oligonucleotides or CRISPR-based approaches. The antisense oligonucleotide therapy QR-110 has shown promising results in early trials, with ongoing phase 3 clinical trials.

Identification of Deep-Intronic Splice Mutations in a Large Cohort of Patients With Inherited Retinal Diseases

Xinye Qian et al.

Summary: This study investigated the contribution of deep-intronic splice variants to inherited retinal disorders by performing whole-genome sequencing on a cohort of 571 patients. Using the SpliceAI tool, six deleterious deep intronic variants were identified, and in vitro experiments showed that mutations with lower prediction scores may result in partial splicing effects.

Beyond nephronophthisis: Retinal dystrophy in the absence of kidney dysfunction in childhood expands the clinical spectrum of CEP83 deficiency

Bram C. F. Veldman et al.

Summary: CEP83 protein plays a crucial role in ciliogenesis and deficiency can lead to ciliopathy. Two unrelated patients with retinitis pigentosa were found to have bi-allelic variants in the CEP83 gene, without nephronophthisis or kidney dysfunction. This study expands the clinical spectrum of CEP83 deficiency and suggests including CEP83 in gene panels for inherited retinal diseases for timely diagnosis.

The need for widely available genomic testing in rare eye diseases: an ERN-EYE position statement

Graeme C. Black et al.

Summary: Rare Eye Diseases (RED) are a significant cause of visual impairment and blindness in Europe, with over 900 disorders in this category, many with a genetic basis. While technological advancements have enabled genetic testing for RED, there is significant variability in access to testing among European countries, with some relying on research-funded laboratories or cross-border testing.

Comparative Natural History of Visual Function From Patients With Biallelic Variants in BBS1 and BBS10

Monika K. Grudzinska Pechhacker et al.

Summary: This study aimed to compare the natural history of visual function change in patients affected with retinal degeneration due to biallelic variants in Bardet-Biedl syndrome genes: BBS1 and BBS10. The results showed that retinal degeneration appeared earlier and was more severe in BBS10 cases compared to BBS1, with visual function decline progressing more rapidly in BBS10 patients.
