Radiation and leukaemia: Which leukaemias and what doses?

Robert Peter Gale

Summary: The cause of most cases of leukaemia is unknown, but exposures to ionizing radiations, certain chemicals, and anti-cancer drugs have been identified as possible causes. Evidence from various sources suggests that ionizing radiations can cause different types of leukaemia, with some types being more sensitive to induction than others. The mechanism by which ionizing radiations cause leukaemia varies for different types. The localization of haemopoietic stem cells in the bone marrow is a topic of discussion.

MiR-483 Promotes Colorectal Cancer Cell Biological Progression by Directly Targeting NDRG2 through Regulation of the PI3K/AKT Signaling Pathway and Epithelial-to-Mesenchymal Transition

Xifeng Sun et al.

Summary: This study revealed the upregulation of miR-483 in colorectal cancer and its prediction of poor prognosis through promoting cell proliferation and invasion. NDRG2 was identified as the target gene of miR-483, and the miR-483/PI3K/AKT pathway and EMT were shown to be involved in cell proliferation and invasion. These findings suggest that miR-483 could be a potential therapeutic target for preventing colorectal cancer.

How I treat chronic-phase chronic myelogenous leukemia

Ellin Berman

Summary: The introduction of imatinib, the first tyrosine kinase inhibitor (TKI) for chronic myelogenous leukemia (CML), revolutionized the treatment of this disease. Subsequent generations of TKIs, such as dasatinib, nilotinib, bosutinib, and ponatinib, have further improved treatment outcomes. Individualized TKI therapy has provided options for patients who do not respond to or cannot tolerate first-line treatment, leading to a significant decrease in the number of patients progressing to advanced stages of the disease. This article reviews newer aspects of clinical care in CML, including the use of bosutinib and asciminib, the cardiac safety of ponatinib, and the consideration of family planning and treatment discontinuation.

Glycogen Storage Disease Phenotypes Accompanying the Perturbation of the Methionine Cycle in NDRG3-Deficient Mouse Livers

Hyun Ahm Sohn et al.

Summary: NDRG3 plays an important role in the regulation of liver cell metabolism, and its loss of function leads to excessive glycogen accumulation, hypoglycemia, elevated liver triglyceride content, and perturbation of the methionine cycle. These findings suggest therapeutic potential for regulating NDRG3 in disorders related to metabolic pathways.

Role of lncSLCO1C1 in gastric cancer progression and resistance to oxaliplatin therapy

Yu-Feng Xiao et al.

Summary: This study found that lncSLCO1C1 is highly expressed in gastric carcinoma (GC) tissue samples and is associated with poor overall survival in patients. lncSLCO1C1 promotes cell proliferation and migration in GC, as well as conferring resistance to oxaliplatin chemotherapy. It acts as a scaffold for the SSRP1/H2A/H2B complex and absorbs miR-211-5p and miR-204-5p, leading to increased expression of SSRP1. The findings highlight the role of lncSLCO1C1 in GC progression.

Proteome dynamics at broken replication forks reveal a distinct ATM-directed repair response suppressing DNA double-strand break ubiquitination

Summary: Cells have evolved an elaborate DNA repair network to ensure accurate and complete DNA replication, with defects in these mechanisms potentially leading to genome instability and therapeutic vulnerabilities. Research shows that ATM signaling stimulates DNA end resection, recruits PLK1, and suppresses the canonical DSB ubiquitination response, providing new insights into how cells orchestrate homologous recombination repair of replication-associated DSBs.

Repression of Wnt/β-catenin signaling by SOX9 and Mastermind-like transcriptional coactivator 2

Abhishek Sinha et al.

Summary: SOX9 promotes turnover of β-catenin independently of the destruction complex and proteasome in mammalian cell culture. This requires the activation of MAML2, which acts as a coactivator for SOX9 in antagonizing the Wnt/β-catenin pathway. This newly identified pathway provides a parallel mechanism for restricting Wnt/β-catenin signaling.
