Hepatic Unsaturated Fatty Acids Are Linked to Lower Degree of Fibrosis in Non-alcoholic Fatty Liver Disease

Michael Friden et al.

Summary: This study investigated the associations between liver fibrosis and fatty acids. Higher levels of TAG 18:1n-9 were associated with lower fibrosis, while higher levels of PL 22:0 were associated with higher fibrosis. These findings may reflect altered fatty acid metabolism.

Branched-Chain Fatty Acids Alter the Expression of Genes Responsible for Lipid Synthesis and Inflammation in Human Adipose Cells

Aleksandra Czumaj et al.

Summary: This study demonstrates for the first time that incubation with iso-BCFA decreases the expression of adipocyte genes associated with lipid metabolism and inflammation, suggesting that changes in the iso-BCFA profile in obese patients may contribute to adipose inflammation and dyslipidemia.

Altered branched-chain α-keto acid metabolism is a feature of NAFLD in individuals with severe obesity

Thomas Grenier-Larouche et al.

Summary: This study found that increased levels of BCKA in the blood and hepatic expression of BCKDK are associated with the presence and severity of NAFLD/NASH in humans. SREBP1 has been identified as a transcriptional regulator of BCKDK.

Production of branched-chain very-long-chain fatty acids by fatty acid elongases and their tissue distribution in mammals

Honoka Tanno et al.

Summary: Branched-chain fatty acids are important components in the meibum lipids secreted from the meibomian glands. Enzymes such as ELOVL1, ELOVL3, and ELOVL7 are responsible for elongating branched-chain acyl-CoAs. Liquid chromatography-tandem mass spectrometry has revealed the predominance of branched-chain lipids in mouse meibomian glands, demonstrating the significance of ELOVL1 in elongating branched-chain VLC acyl-CoAs in vivo.

Pathophysiological communication between hepatocytes and non-parenchymal cells in liver injury from NAFLD to liver fibrosis

Santosh Kumar et al.

Summary: Non-alcoholic fatty liver disease (NAFLD) is a multifactorial disease with a spectrum of pathological conditions, involving liver cell injury and signaling pathways such as lipotoxicity, oxidative stress, inflammation, etc., ultimately leading to liver regeneration and fibrogenesis. Targeting specific liver cell types using nanotechnology shows potential as a therapeutic strategy for NASH and liver fibrosis.

The Role of Fatty Acids in Non-Alcoholic Fatty Liver Disease Progression: An Update

Aleksandra Hliwa et al.

Summary: Non-alcoholic fatty liver disease (NAFLD) is a major global public health problem characterized by alterations in hepatic lipid metabolism that can lead to serious liver complications. Mass spectrometry-based lipidomics platforms allow for in-depth analysis of lipid changes in NAFLD patients, focusing on alterations in long-chain and short-chain fatty acids levels as well as the expression of genes encoding lipid interconversion enzymes in the liver.

The role of hepatic lipid composition in obesity-related metabolic disease

Scott A. Willis et al.

Summary: Studies have shown a close relationship between excessive hepatic lipid accumulation and metabolic dysfunction, with the composition of hepatic lipids playing a more direct role in affecting metabolic abnormalities. Elevated saturated and monounsaturated hepatic lipids and decreased polyunsaturated hepatic lipids are associated with metabolic derangements, such as insulin resistance.
