Transcription associated cyclin-dependent kinases as therapeutic targets for prostate cancer

Theodora A. Constantin et al.

Summary: Transcriptional deregulation is a characteristic feature of various cancer types. In metastatic castration-resistant prostate cancer, the addiction of transcriptional activity to the androgen receptor is maintained in most patients. Transcription associated cyclin-dependent kinases (tCDKs), a group of protein kinases, have shown great potential as therapeutic targets for prostate cancer and other solid tumors.

Somatic Mutations in Core Spliceosome Components Promote Tumorigenesis and Generate an Exploitable Vulnerability in Human Cancer

Claudio Sette et al.

Summary: High throughput exome sequencing has revealed recurrent cancer-associated mutations in spliceosomal components, which promote tumor phenotype and make cancer cells vulnerable to splicing-targeting approaches. Thus, understanding the mechanisms and roles of abnormal splicing in tumor metabolism could lead to the development of novel tumor-targeting drugs.

The transcriptional terminator XRN2 and the RNA-binding protein Sam68 link alternative polyadenylation to cell cycle progression in prostate cancer

Marco Pieraccioli et al.

Summary: The selection of polyadenylation signals and alternative polyadenylation (APA) play important roles in gene expression regulation. This study uncovers a mechanism of APA regulation involving the interaction between XRN2 exonuclease and Sam68 RNA-binding protein. The increased expression of Sam68 in prostate cancer, promoted by MYC, leads to the usage of suboptimal proximal polyadenylation signals and enhances cell cycle-related gene translation. This finding suggests that the Sam68/XRN2-driven APA program may be a potential target for therapeutic strategies.

The oncogenic kinase NEK2 regulates an RBFOX2-dependent pro-mesenchymal splicing program in triple-negative breast cancer cells

Chiara Naro et al.

Summary: The study reveals an extensive splicing program modulated by NEK2 involving splice variants that confer an invasive phenotype to TNBCs, with potential as therapeutic targets. NEK2-sensitive exons could serve as markers for the TNBC subtype and have prognostic value.

Oncogenic dysregulation of pre-mRNA processing by protein kinases: challenges and therapeutic opportunities

Chiara Naro et al.

Summary: Alternative splicing and polyadenylation play crucial roles in the diversification of the human transcriptome and proper gene expression. Dysregulation of these processes can contribute to oncogenesis and treatment resistance in cancer. Protein kinase families such as SRPK, CLK, and CDK are key players in fine-tuning pre-mRNA processing mechanisms, and their inhibition has shown anticancer effects in cancer cells.
