Summary: This study reveals the important role of the cellular RNA-binding protein PCBP1 in the hepatitis C virus (HCV) life cycle. Knockdown of PCBP1 leads to a decrease in viral RNA accumulation but an increase in extracellular viral titers. PCBP1 knockdown does not directly affect viral entry, translation, RNA stability, or replication, but results in an overall increase in infectious particle secretion. Blocking virus secretion can partially restore the viral RNA accumulation decreased by PCBP1 knockdown.

High-Throughput Analysis Reveals miRNA Upregulating α-2,6-Sialic Acid through Direct miRNA-mRNA Interactions

Summary: Chemical biology has revealed the significance of sialic acids as important signals in physiology and disease. miRNAs, noncoding RNAs that downregulate protein expression, have been shown to play a role in regulating glycosylation. Contrary to expectations, our study found that the majority of miRNAs upregulate ST6GAL1 and alpha-2,6-sialylation in cancer cells, while miRNAs that regulate ST6GAL2 were predominantly downregulatory. This challenges previous assumptions about miRNA and suggests a potential pathway for explaining the dysregulation of sialylation observed in cancer and other diseases.
