A 2'-phosphouridine modification at position 47 of tRNAs in thermophilic archaea was identified, which confers thermal stability and nuclease resistance to tRNAs. The modification is mediated by the enzyme ArkI and dephosphorylated by the enzyme KptA, and it fine-tunes the structural rigidity of tRNAs under extreme environmental conditions.
