Roles of homologous recombination in response to ionizing radiation-induced DNA damage

Jac A. Nickoloff et al.

Summary: Ionizing radiation induces DNA lesions, including DSBs, which can lead to genome alterations and carcinogenesis. DSBs are repaired by NHEJ or HR, with HR playing a crucial role in cellular responses to radiation. HR may be an exploitable target for cancer radiotherapy.

A pan-cancer compendium of chromosomal instability

Ruben M. Drews et al.

Summary: This study evaluates the extent, diversity, and origin of chromosomal instability (CIN) in tumors of different cancer types and provides a compendium of copy number signatures that characterize specific types of CIN. These signatures predict drug response and identify new drug targets, offering guidance for future CIN research.

Biochemical properties of fission yeast homologous recombination enzymes

Hideo Tsubouchi et al.

Summary: Homologous recombination (HR) is a universal phenomenon found from viruses to humans, with essentially the same mechanisms in humans and simple unicellular eukaryotes like yeast. The highly diverged yeast species, Saccharomyces cerevisiae and Schizosaccharomyces pombe, have been exceptionally useful in understanding eukaryotic HR by providing unique biological insights and complementing each other. This review focuses on comparing the features of HR mechanisms in S. pombe with other model organisms, with emphasis on the biochemical characterization of HR mechanisms revealed using S. pombe proteins.

The Safe Path at the Fork: Ensuring Replication-Associated DNA Double-Strand Breaks are Repaired by Homologous Recombination

Jac A. Nickoloff et al.

Summary: Cells need to accurately replicate and segregate DNA to prevent cancer, but replication forks can encounter obstacles leading to replication stress. Cells respond by activating DNA damage response pathways to delay cell cycle progression, stimulate repair and replication fork restart, or induce apoptosis. Resection of single-ended double-strand breaks (seDSBs) and accurate fork restart by homologous recombination are crucial in maintaining genome stability.

Structural Chromosome Instability: Types, Origins, Consequences, and Therapeutic Opportunities

Sebastian Omar Siri et al.

Summary: Chromosome instability (CIN) involves increased numerical and structural changes in chromosomes, playing a pivotal role in tumor progression and potential therapeutic targeting. The review delves into the origins of structural CIN, cellular mechanisms, relationships between different CIN phenotypes, and the consequences of structural CIN in cells. The discussion also explores strategies to either prevent or amplify CIN for tumor treatment.
