An efficient LC-MS method for isomer separation and detection of sugars, phosphorylated sugars, and organic acids

Summary: This study developed a sensitive liquid chromatography-mass spectrometry method that selectively enhanced the sensitivity of 27 central metabolites, especially phosphorylated sugars, in a single run. By combining hydrophilic interaction chromatography with anion exchange chromatography, the method improved the peak areas of phosphorylated sugars in plant tissues. Additionally, the method was applied to investigate metabolism in high lipid-producing tobacco leaves.

Metabolic flux analysis of the non-transitory starch tradeoff for lipid production in mature tobacco leaves

Summary: The metabolic plasticity of tobacco leaves was demonstrated by generating transgenic plants with high lipid accumulation. Research showed that foliar lipids increased gradually with plant development, while starch content decreased, indicating a metabolic carbon tradeoff between starch and lipids. Additionally, the contribution of NADP-malic enzyme to plastidic pyruvate production for lipid synthesis was highlighted as a key factor in the successful leaf lipid engineering efforts in tobacco.

Expression of AtWRI1 and AtDGAT1 during soybean embryo development influences oil and carbohydrate metabolism

The results showed that the expression of these genes did not increase the total fatty acid content, but led to changes in the oil and carbohydrate composition. Suppression of SDP1 Improves Soybean Seed Composition by Increasing Oil and Reducing Undigestible Oligosaccharides

Summary: This study demonstrates that suppressing lipase activity in soybean seeds can increase fatty acid content and decrease the presence of undigestible oligosaccharides. The upregulation of lipase genes during late seed development decreases fatty acid content and increases oligosaccharide levels. Suppressing these genes leads to higher oil levels and larger seeds without negatively impacting protein content.

Effects of type I Diacylglycerol O-acyltransferase (DGAT1) genes on soybean (Glycine max L.) seed composition

Summary: This study demonstrated the importance of DGAT1 genes in determining seed compositions in soybean, by downregulating three isoforms which led to a decrease in seed oil concentration and an increase in seed protein concentration, as well as changes in fatty acid compositions. This study demonstrates the utility of microalgal cells in studying acyl-ACP dynamics for insights into plant FA biosynthesis regulation and oil enhancement strategies.
