NRAMP6 and NRAMP1 cooperatively regulate root growth and manganese translocation under manganese deficiency in Arabidopsis

Lun Li et al.

Summary: NRAMP6 plays an important role in manganese (Mn) nutrition and long-distance signaling between roots and shoots under Mn deficient conditions. It cooperates with NRAMP1 to regulate root growth and Mn translocation. The protein expression and localization of NRAMP6 are regulated by Mn deficiency, and it can rescue the defective growth of a yeast mutant deficient in endomembrane Mn transport.

A Golgi-Located Transmembrane Nine Protein Gene TMN11 Functions in Manganese/Cadmium Homeostasis and Regulates Growth and Seed Development in Rice

He Li et al.

Summary: Metal transporters play crucial roles in plant nutrition, development, and metal homeostasis. In this study, a new transmembrane nine (TMN) family gene, TMN11, was identified as a Mn transport protein localized to the cis-Golgi endomembrane in rice. OsTMN11 was ubiquitously expressed over the lifespan of rice and strongly upregulated in young rice under excess Mn(II)/Cd(II) stress. Functional analysis revealed that OsTMN11 functions in a similar manner to ScPMR1 in yeast, mediating Mn efflux. Knockdown of OsTMN11 compromised rice growth and seed development under excessive Mn and Cd conditions.

Duplication of NRAMP3 Gene in Poplars Generated Two Homologous Transporters with Distinct Functions

Mathieu Pottier et al.

Summary: The study finds that there are two copies of NRAMP3 metal transporters in the poplar genome, one of which retains the original function of releasing metals stored in vacuoles in Arabidopsis, while the other copy has acquired a new function in controlling the cell-to-cell transport of manganese in the Trans-Golgi Network. This research provides a clear case of neofunctionalization through a change in the subcellular localization of a metal transporter and evidence for the involvement of the secretory pathway in the cell-to-cell transport of manganese.

Manganese is a physiologically relevant TORC1 activator in yeast and mammals

Raffaele Nicastro et al.

Summary: This research reveals that elevated levels of cytoplasmic manganese stimulate the activity of TORC1 kinase, regulating cell growth and metabolism. Manganese activates TORC1 more efficiently than magnesium and can control autophagy and mitochondrion-to-nucleus retrograde signaling. These findings are significant for understanding the role of manganese in human diseases.

Golgi-localised manganese transporter PML3 regulates Arabidopsis growth through modulating Golgi glycosylation and cell wall biosynthesis

Chang-Hong Yang et al.

Summary: PML3 in Arabidopsis is involved in manganese transport, Golgi glycosylation, and cell wall biosynthesis under manganese-deficient conditions. Double mutations of PML3 and NRAMP2 showed improved plant growth under manganese deficiency, indicating opposite roles of PML3 and NRAMP2 in regulating Golgi manganese levels.

Manganese triggers phosphorylation-mediated endocytosis of the Arabidopsis metal transporter NRAMP1

Loren Castaings et al.

Summary: NRAMP1 transporter mediates plant survival of manganese deficiency by controlling manganese entry into root cells. The phosphorylation of NRAMP1 triggers its endocytosis and membrane distribution, regulating plant tolerance to manganese toxicity. Interrupting NRAMP1 membrane cycling leads to increased manganese toxicity, challenging the belief that manganese is only marginally toxic to plants.

A Golgi-localized manganese transporter functions in pollen tube tip growth to control male fertility in Arabidopsis

Bin Zhang et al.

Summary: A Golgi-localized Mn transporter named PML3 is essential for rapid cell elongation in young tissues like the pollen tube, and its loss-of-function mutation results in reduced seed yield due to decreased male fertility. The lack of pectin deposition in the pollen tube may cause bursting and slow elongation, highlighting the importance of PML3 in Mn homeostasis for metabolic and trafficking processes required for pectin deposition in rapidly elongating cells.
