The impact of insomnia on frailty and the hallmarks of aging

Catarina Carvalhas-Almeida et al.

Summary: Despite age-related changes in sleep, a high percentage of older adults experience sleep dissatisfaction and chronic insomnia. Chronic insomnia, the most common sleep disorder worldwide, is expected to increase due to aging and obesity. This can lead to adverse health outcomes, promoting physical and cognitive decline and aggravating frailty in older adults. The link between chronic insomnia, hallmarks of aging, and frailty is discussed in this review, along with the potential effects of insomnia therapeutic approaches on cellular aging.

Melatonin attenuates reactive astrogliosis and glial scar formation following cerebral ischemia and reperfusion injury mediated by GSK-3 beta and RIP1K

Nuttapong Yawoot et al.

Summary: Melatonin has long-term benefits on cerebral damage after ischemia and reperfusion injury, as it reduces infarct volume, neuronal cell death, reactive astrogliosis, and glial scar formation, while promoting axonal regeneration and neurobehavioral recovery. By inhibiting the activities of GSK-3 beta and RIP1K, melatonin effectively counteracts the deleterious effects due to astrocyte responses and improves functional recovery during the chronic phase of cerebral ischemia and reperfusion injury.

Chronic microglial inflammation promotes neuronal lactate supply but impairs its utilization in primary rat astrocyte-neuron co-cultures

Yi Wang et al.

Summary: Neuronal activity is closely linked to energy metabolism, and the mechanisms underlying inflammation-induced neuronal injury are not fully understood. This study revealed that chronic inflammation affects the levels and metabolism of lactate in astrocytes and neurons, leading to increased lactate supply to neurons but impaired lactate oxidation. Additionally, chronic inflammation disrupts the neuronal cytoskeleton.

Microglia modulates hippocampal synaptic transmission and sleep duration along the light/dark cycle

Giorgio Corsi et al.

Summary: Microglia play a crucial role in regulating sleep by adapting their cx3cr1 expression level and modulating synaptic activity in a phase-dependent manner. The findings suggest the importance of microglial cells in the homeostasis of cerebral parenchyma and the regulation of the sleep/wake cycle.

Astrocyte-derived adenosine excites sleep-promoting neurons in the ventrolateral preoptic nucleus: Astrocyte-neuron interactions in the regulation of sleep

In-Sun Choi et al.

Summary: This study reveals a novel mechanism involving astrocyte-neuron interactions in sleep regulation, where ATP derived from astrocytes is converted into adenosine by TNAP. Adenosine inhibits non-sleep-promoting neurons and excites sleep-promoting neurons, thereby affecting sleep-wake cycle. This finding has important implications for understanding the physiological basis of sleep regulation.

BRAINMECHANISMS OF INSOMNIA: NEW PERSPECTIVES ON CAUSES AND CONSEQUENCES

Eus J. W. Van Someren

Summary: Insomnia is the second most common mental disorder, yet progress in understanding its underlying neurobiological mechanisms has been limited. This review explores the definition, prevalence, and susceptibility factors of insomnia, including genetic variants, early life stress, major life events, and brain structure and function. The proposed model suggests that in individuals vulnerable to developing insomnia, the locus coeruleus may be more sensitive to salience network input, leading to a spiral of hyperarousal and impaired sleep.

The Regulation of Drosophila Sleep

Orie T. Shafer et al.

Summary: Sleep is crucial for brain function across different animal species, and genetic studies in fruit flies have provided detailed insights into the genes and neural circuits that regulate sleep. The research shows that the mechanisms of sleep regulation are largely conserved from flies to mammals, and that sleep is not a single state but consists of multiple physiological and behavioral states influenced by the environment and life history.

Brain glycogen metabolism: A possible link between sleep disturbances, headache and depression

J-M Petit et al.

Summary: The functions of sleep and its links with neuropsychiatric diseases have long been questioned. The role of glycogen in sleep is crucial, as inadequate turnover may be one of the mechanisms linking migraine, mood disorders, and sleep. Sleep deprivation-induced transcriptional changes may trigger migraine by reducing glycogen availability, leading to susceptibility to cortical spreading depolarization. Additionally, chronic stress affecting brain glycogen turnover through various pathways may contribute to mood disorders and sleep disturbances.

Summary: In this study, changes in astrocyte Ca2+ levels in different brain regions of mice during sleep/wakefulness states were examined using fiber photometry. The research showed that Ca2+ levels in astrocytes decrease during REM sleep, increase after wakefulness onset, and vary among brain regions during NREM sleep. The study implies that astrocytes may play diverse physiological roles in sleep based on the differences in Ca2+ dynamics observed across brain regions.
