Blockage of HCN Channels Inhibits the Function of P2X Receptors in Rat Dorsal Root Ganglion Neurons

Summary: This study aimed to investigate the modulation of HCN channel activity on the expression and function of P2X2 and P2X3 receptors. The results showed that intrathecal administration of HCN channel blocker ZD7288 significantly reduced mechanical hyperalgesia and the increased expression of P2X2 and P2X3 receptors in rats with chronic constriction injury of sciatic nerve. Furthermore, it was found that the opening of HCN channels could enhance the expression and function of P2X2 and P2X3 receptors via the cAMP-PKA signaling pathway. These findings provide important insights into the mechanism of nerve injury-induced pain hypersensitivity.

Meclizine and Metabotropic Glutamate Receptor Agonists Attenuate Severe Pain and Ca2+ Activity of Primary Sensory Neurons in Chemotherapy-Induced Peripheral Neuropathy

Summary: Chemotherapy-induced peripheral neuropathy (CIPN) is a common side effect of chemotherapy and can cause severe neuropathic pain. In this study, the researchers found that activating mGluR8 and Group II mGluR, as well as using meclizine, could reduce the pain and weight loss caused by cisplatin-induced CIPN. These findings suggest that these drugs may be promising new treatment options for CIPN.
