A systematic review and meta-analysis of the effectiveness and adverse events of gabapentin and pregabalin for sciatica pain

Maria Soledad Gimenez-Campos et al.

Summary: This systematic review found lack of effectiveness of pregabalin and gabapentin in managing sciatica pain and disability. Therefore, routine clinical use of these medications cannot be supported.

Symptom-based pharmacotherapy for neuropathic pain related to spinal disorders: results from a patient-based assessment

Hideaki Nakajima et al.

Summary: Existing guidelines recommend an updated therapeutic algorithm for chronic neuropathic pain, but individualized pharmacotherapeutic management based on pain phenotypes may achieve higher efficacy. This retrospective study evaluated the efficacy of medications based on neuropathic pain phenotypes and proposed symptom-based pharmacotherapy. The findings could contribute to better strategies for symptom-based pharmacotherapeutic management.

Decoding gene expression signatures in mice trigeminal ganglion across trigeminal neuropathic pain stages via high-throughput sequencing

Fei-Fei Xu et al.

Summary: This study assessed the transcriptome of the trigeminal ganglion during the development and maintenance phases of trigeminal neuropathic pain (TNP). The results revealed altered gene expression profiles and identified potential hub genes and pathways, such as the MAPK pathway, that may serve as targets for TNP treatment.

Sex-differences in network level brain dynamics associated with pain sensitivity and pain interference

Junseok A. Kim et al.

Summary: Pain sensitivity and pain interference are closely related to functional connectivity within and across brain networks, with gender differences observed in these relationships. Women exhibit higher static functional connectivity, while men have higher dynamic functional connectivity, and the impact of pain sensitivity and interference on functional connectivity varies across different frequency bands.

Sex- and age-specific genetic analysis of chronic back pain

Maxim B. Freidin et al.

Summary: Sex differences exist in chronic back pain (cBP), with females usually experiencing higher morbidity, severity, and poorer treatment response. Genetic factors may play a role in an age-specific manner, with a high genetic correlation between the sexes in cBP.

Animal models of pain: Diversity and benefits

Cynthia Abboud et al.

Summary: Chronic pain is a major health problem and studying animal models that recapitulate diverse pain symptoms is crucial for understanding pain mechanisms. These models reproduce different pain phenotypes, allowing for examination of multidimensional aspects of pain and providing insights into the cellular and molecular basis of pain conditions. The validity of these models is based on their ability to mimic human diseases and predict treatment outcomes.

Pharmacological modulation of brain activation to non-noxious stimulation in a cynomolgus macaque model of peripheral nerve injury

Aldric Hama et al.

Summary: In vivo neuroimaging in a cynomolgus macaque model of unilateral peripheral neuropathy revealed changes in CNS neurons following nerve injury and allowed for the differentiation between the clinical efficacy of analgesic drugs, such as pregabalin, and drugs with less clinical efficacy, like aprepitant.
