Integrated Proteome and Phosphoproteome Analyses Reveal Early- and Late-Stage Protein Networks of Traumatic Brain Injury

Summary: In this study, differentially expressed proteins (DEPs) and differentially expressed phosphoproteins (DEPPs) in the early and late stages of TBI were identified using TMT labeling, Phos affinity enrichment, and high-resolution LC-MS/MS analysis. Integrative analyses revealed the different and similar pathophysiologic mechanisms between the early and late stages, providing insights into useful biomarkers and underlying mechanisms in TBI and its sequelae.

This study used functional MRI to examine the impact of Ras/MAPK pathogenic variants on brain organization and cognitive phenotypes in children with Noonan syndrome (NS). The results showed hyperconnectivity within canonical brain networks in the NS group, which may represent an intermediate phenotype between Ras/MAPK pathogenic variants and cognitive phenotypes.

Summary: Repetitive mild traumatic brain injury (r-mTBI) is the most widespread type of brain trauma worldwide, leading to long-lasting pathological and molecular changes that increase the risk of chronic neurodegenerative diseases. Our study identified numerous proteins and phosphoproteins that remain continuously dysregulated from 3 to 24 weeks post-TBI, which could serve as effective therapeutic targets for sub-acute and chronic stages of post r-mTBI. The translational relevance of our mouse model of r-mTBI was supported by the identification of common proteins and pathways with autopsied human samples of Chronic Traumatic Encephalopathy (CTE) patients, highlighting the potential for successful translation of therapeutic targets.
