High-speed high-resolution data collection on a 200 keV cryo-TEM

Molecular basis for stereoselective transport of fenoterol by the organic cation transporters 1 and 2

Summary: Stereoselectivity plays an important role in drug membrane transport, but its impact is still not well understood. This study investigated the stereoselective effects of fenoterol enantiomers in cellular uptake, and proposed possible molecular explanations through computational modeling and experimental validation. The results suggest that fenoterol enantiomers bind to distinct binding sites in OCT1 and OCT2, leading to the observed stereoselectivity differences.

Recent advances on the inhibition of human solute carriers: Therapeutic implications and mechanistic insights

Summary: Solute carriers (SLCs) are membrane proteins that mediate the transport of hydrophilic molecules across cell membranes. Despite their important roles in human biology, they are underexplored as therapeutic targets. This review discusses successful drug cases that inhibit SLCs and explores the therapeutic potential of certain human SLCs. The review also highlights recent structural studies that provide insights into the inhibition mechanisms of two clinically relevant human major facilitator super-family (MFS) transporters.

Amino acids in transmembrane helix 1 confer major functional differences between human and mouse orthologs of the polyspecific membrane transporter OCT1

Summary: This study analyzed the differences between human and mouse OCT1 orthologs and identified nonconserved amino acids Cys36 and Phe32 as determinants of OCT1 polyspecificity. It also found that the second phenol ring in Cys36 is essential for the affinity of fenoterol. This is the first study to report single amino acids as determinants of OCT1 polyspecificity.
