Clonally expanded B cells in multiple sclerosis bind EBV EBNA1 and GlialCAM

Tobias Lanz et al.

Summary: Multiple sclerosis (MS) is an autoimmune disease where autoreactive lymphocytes attack the central nervous system. Epstein-Barr virus (EBV) infection has been linked to MS, with molecular mimicry between EBV nuclear antigen 1 (EBNA1) and central nervous system protein GlialCAM. EBNA1 immunization exacerbates disease in MS mouse models and anti-EBNA1 and anti-GlialCAM antibodies are prevalent in MS patients, providing a mechanistic link between MS and EBV and guiding the development of new MS therapies.

Human Leukocyte Antigen Association Study Reveals DRB1*04:02 Effects Additional to DRB1*07:01 in Anti-LGI1 Encephalitis

Vicente Peris Sempere et al.

Summary: The study found that HLA alleles DRB1*07:01 and DQA1*02:01 are associated with anti-LGI1 encephalitis, independent of variation at DRB3 and DQB1 loci. Additionally, the DRB1*04:02 allele is also independently associated with the disease and lower age at onset. These findings provide evidence for secondary effects within the HLA locus that correlate with clinical phenotypes in anti-LGI1 encephalitis.

Clinical and Prognostic Value of Immunogenetic Characteristics in Anti-LGI1 Encephalitis

Sergio Muniz-Castrillo et al.

Summary: Anti-LGI1 antibodies are associated with a form of limbic encephalitis strongly linked to HLA-DRB1*07:01, but some patients lack these antibodies in their cerebrospinal fluid. Patients without CSF LGI1-Abs show no clinical differences, suggesting a lesser production of LGI1 antibodies in these cases. The HLA association is biased towards age and sex, indicating subtle differences in immune response. Long-term outcome is mostly dependent on demographic characteristics and the intensity of the intrathecal synthesis.
