Single-cell transcriptomic analysis of human colonic macrophages reveals niche-specific subsets

Summary: Macrophages in the intestine play important roles in tissue homeostasis and immune response. They are composed of different subsets with distinct functions. Colonic lamina propria macrophages (LpMs) include subsets with proinflammatory properties and subsets with high antigen-presenting and phagocytic capacity. Muscularis macrophages (MMs) differentiate along two trajectories, one associated with immune activation and angiogenesis, and the other associated with neuronal homeostasis. The survival, migration, transcriptional reprogramming, and niche-specific localization of LpMs and MMs are controlled by interactions with tissue-resident cells and key transcription factors.

Macrophage depletion systemically and/or exclusively in the colon improves glucose metabolism. This study highlights the importance of colonic macrophage subpopulations in regulating glucose homeostasis and suggests that pharmacological targeting of these macrophages may be a potential therapy for obesity-related glycemic control.

Summary: Dysfunctional resolution of intestinal inflammation and altered mucosal healing are critical in the pathogenesis of inflammatory bowel disease (IBD). In this study, we identified a subset of PTGER4-expressing intestinal macrophages with mucosal healing properties in humans and mice. Mechanistically, increased mucosal levels of PGE(2) trigger the secretion of CXCL1 in PTGER4(+) macrophages via MAPKs, promoting epithelial cell differentiation and proliferation in colitis. Targeting macrophages with liposomes loaded with an MAPK agonist could restore defective epithelial regeneration and promote mucosal healing, offering potential therapeutic targets for patients with IBD.
