Synaptic Damage and Its Clinical Correlates in People With Early Huntington Disease A PET Study

Aline Delva et al.

Summary: The study found synaptic damage in early HD, starting in the striatum in the premanifest phase and spreading extensively to extrastriatal regions related to motor impairment in the early manifest phase. C-11-UCB-J PET is more sensitive than F-18-FDG-PET for detecting extrastriatal changes in early HD.

Healthy brain aging assessed with [18F]FDG and [11C]UCB-J PET

Katrine B. Andersen et al.

Summary: The study using PET imaging found widespread cortical reduction of synaptic density in older adults, but the reduction did not persist after corrections. Regional differences in glucose metabolism and synaptic density were observed, suggesting a non-uniform relationship between the two variables in the human brain and implications for neuroenergetics.

Summary: We conducted a comparison study on the synaptic density and metabolism in AD and CN participants using [C-11]UCB-J PET and [F-18]FDG PET, finding a similar reduction in the medial temporal lobe but a discordant reduction in neocortical regions, with higher correlations between synaptic density and metabolism in the former.

Reduced Synaptic Density in Patients with Lewy Body Dementia: An [11C]UCB-J PET Imaging Study

Summary: This study evaluated synaptic density in PD and dementia patients using in vivo PET imaging and found widespread cortical reduction in DLB/PDD subjects. The results suggest that synaptic density loss contributes to cognitive impairment in nPD and DLB/PDD.

Synaptic Density and Neuronal Metabolic Function Measured by Positron Emission Tomography in the Unilateral 6-OHDA Rat Model of Parkinson's Disease

Summary: The study investigated [C-11]UCB-J binding and [F-18]FDG uptake in the CSTC circuit following a unilateral dopaminergic lesion in rats, showing lower binding and uptake in the ipsilateral striatal regions compared to the contralateral regions. Differential changes between hemispheres for [C-11]UCB-J and [F-18]FDG outcomes were evident, especially in cortical regions. The results suggest that combined [C-11]UCB-J and [F-18]FDG scans could provide a more comprehensive understanding of heterogeneous cerebral changes in neurodegenerative disorders.
