Summary: Beta-cyclodextrins and beta-CD-containing polymers are potential candidates for the treatment of cholesterol-related diseases. In this study, we modified polyrotaxanes (PRXs) with sulfobetaine and amine groups to enhance their solubility and functionality. The modified PRXs showed high cholesterol-reducing ability and biocompatibility, making them promising for therapeutic applications. This research provides insights for the design of materials with both bioinert and bioactive properties.

Activated cholesterol metabolism is integral for innate macrophage responses by amplifying Myd88 signaling

Summary: Recent studies have shown a close relationship between cellular metabolism and the regulation of immune cells. This study reveals that cholesterol metabolism plays an integral role in innate immune responses of macrophages. Cholesterol accumulation in endosomes and lysosomes is crucial for amplification of Myd88 signaling, which is involved in inflammatory macrophage activation. The supramolecular compound PRX is able to inhibit inflammatory macrophage activation by regulating cholesterol trafficking and efflux. These findings highlight the importance of cellular cholesterol metabolism in innate activation of macrophages and suggest its potential as a therapeutic target for inflammatory diseases.
