Interaction of a Homologous Series of Amphiphiles with P-glycoprotein in a Membrane Environment-Contributions of Polar and Non-Polar Interactions

Maria Joao Moreno et al.

Summary: The transport of drugs by efflux transporters plays a crucial role in drug resistance development by cancer cells and pathogens. In this study, the interaction between a series of fluorescent amphiphiles and the efflux transporter P-glycoprotein (P-gp) was explored. The results showed that the binding parameters obtained were apparent and a new formalism was proposed to better understand the binding affinity of amphiphiles with P-gp. It was found that the polar interactions are essential for this interaction, and an increase in lipophilicity and amphiphilicity enhanced the association with the lipid bilayer, inhibiting the function of P-gp.

P-glycoprotein (ABCB1)- weak dipolar interactions provide the key to understanding allocrite recognition, binding, and transport

Anna Seelig et al.

Summary: P-glycoprotein is an ATP binding cassette transporter that facilitates the movement of compounds across membranes. This study investigates the thermodynamics of compound binding and ATP hydrolysis by P-glycoprotein. The findings suggest a new model for compound transport by P-glycoprotein, taking into account the evolutionary optimization and the interactions within the membrane. Weak dipolar interactions are found to drive compound recognition, binding, and transport. The proposed unidirectional transport cycle is consistent with membrane biophysics.

Summary: This study investigates the dynamic consequences of substrate and inhibitor binding to mouse P-gp using hydrogen-deuterium exchange mass spectrometry. The results reveal that these drugs affect the dynamics of specific regions, leading to stimulation of ATP hydrolysis. However, the effects of drug binding diverge in the post-hydrolysis state.

Analysis of the Equilibrium Distribution of Ligands in Heterogeneous Media-Approaches and Pitfalls

Maria Joao Moreno et al.

Summary: This review article discusses various methods and formalisms for analyzing the equilibrium distribution of ligands in the presence of proteins, lipid membranes, or both, with special attention to common pitfalls and validity limits of different approaches. The analysis of ligand binding through Stern-Volmer plots of protein fluorescence quenching is emphasized for its widespread use in characterizing interactions. Different systems of increasing complexity are considered, from proteins with single to multiple binding sites to membrane proteins in lipid bilayers.
