Ameliorative effects of bromelain on aluminum-induced Alzheimer's disease in rats through modulation of TXNIP pathway

Summary: Alzheimer's disease (AD) is linked to brain insulin resistance, and this study explored the roles of TXNIP, P-IRS-1, and PI3K in AD pathogenesis. The study found that bromelain showed similar effects to donepezil in improving cognitive functions, anxiety, and depression levels in an AD rat model. Bromelain and donepezil both ameliorated neuronal degeneration and amyloid plaque deposition. Acetylcholine esterase levels were decreased by bromelain and donepezil treatment. The study also revealed the modulatory effect of bromelain on the TXNIP/P-IRS-1/PI3K axis in AD.

Discovery of New 1,3,4-Oxadiazoles with Dual Activity Targeting the Cholinergic Pathway as Effective Anti-Alzheimer Agents

Summary: This study presents the design, synthesis, and evaluation of a new series of compounds based on a 5-pyrid-3-yl-1,3,4-oxadiazole scaffold as potential therapeutically effective agents against Alzheimer's disease (AD). The synthesized compounds displayed excellent in vitro enzyme inhibitory activity against major AD disease-modifying targets. Compound 5e was found to be the best dual inhibitor of acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE) among all the compounds evaluated, surpassing the FDA-approved inhibitor rivastigmine. In vivo and ex vivo testing of compound 5e further demonstrated its significant AD-biotargeting effects.

Summary: This study investigated the chemical composition, antioxidant, alpha-glucosidase, alpha-amylase, and cholinesterase inhibitory activities of wormwood and rosemary essential oils. Molecular docking tools were used to explain the interaction between chemical constituents and enzymes. The results showed that rosemary oil had higher antioxidant and cholinesterase inhibitory activities, while wormwood oil was more effective against antidiabetic inhibitory enzymes. Molecular modeling verified the strong binding affinity of certain chemical compounds from rosemary with the studied enzymes. These findings contribute to the understanding of the biological activities of rosemary and wormwood and their potential applications in the food and pharmaceutical industries.

Additionally, the oregano extract exhibited high antioxidant activity.
