Crystallization Propensity of Amorphous Pharmaceuticals: Kinetics and Thermodynamics

Summary: The study used four model compounds with nearly identical glass transition temperatures to investigate the effects of thermodynamics and molecular mobility on their crystallization propensities. Despite similar molecular mobility, the significant difference in crystallization propensities among the compounds was attributed to variations in the thermodynamic barriers.

Indomethacin: The Interplay between Structural Relaxation, Viscous Flow and Crystal Growth

Summary: Non-isothermal differential scanning calorimetry (DSC) was used to investigate the effects of particle size and heating rate on the structural relaxation, crystal growth, and decomposition kinetics of amorphous indomethacin. The results showed that the kinetics of structural relaxation and decomposition were independent of particle size, while they depended on the heating rate. The crystal growth kinetics determined by DSC played a dominant role in the macroscopic amorphous-to-crystalline transformation. The study also discussed the correlations between macroscopic and microscopic crystal growth processes, as well as the relationship between viscous flow and structural relaxation motions.

Stabilizing the amorphous form through methods like solid dispersion and co-amorphous can enhance the dissolution and bioavailability of nutraceuticals. This review presents an application of amorphous systems in delivering poorly soluble nutraceuticals to overcome their pharmacokinetic limitations.

Single-curve multivariate kinetic analysis: Application to the crystallization of commercial Fe-Si-Cr-B amorphous alloys

Summary: The crystallization behavior of three commercial Fe-Si-Cr-B amorphous alloys was investigated through various techniques including differential scanning calorimetry, optical and scanning electron microscopies, and X-ray diffraction analysis. The complex crystallization kinetics was described using a transformation mechanism of multiple overlapping independent processes following the autocatalytic Sestak-Berggren model. A newly developed method of single-curve multivariate kinetic analysis (sc-MKA) accurately predicted the degrees of conversion achieved during annealing and growth rates during heating for certain alloys.

Crystallization of glasses - When to use the Johnson-Mehl-Avrami kinetics?

Summary: The study tested the applicability of the JMA model based on the universal criterion of the kinetic peak asymmetry and determined the correlation between the simulated crystallization peaks and the JMA description. Results showed that the JMA model provided accurate kinetic predictions with high correlation coefficients within specific intervals of conversion degrees.
