Summary: New derivatives, 3-Nor-4 beta-friedelan-24-ol (1) and 3-decyl-6,7,8-trimethoxy-2H,5H-furo[4,3,2-de]isochromene-2,5-dione (4), were isolated and characterized from Rinorea oblongifolia fruits, along with other known compounds. The enzyme inhibitory activities of these compounds were evaluated, showing good activity against acetylcholinesterase (AChE), butyrylcholinesterase (BChE), alpha-glucosidase, urease and tyrosinase. The results suggest that Rinorea oblongifolia and its constituents have potential therapeutic applications for enzymatic disorders.

Influence of Side Chain Conformation on the Activity of Glycosidase Inhibitors

Summary: The conformation of substrate side chain affects reactivity in glycosylation and glycoside hydrolysis reactions and is restricted during catalysis by many enzymes. This study demonstrates that gluco and manno iminosugars can be restricted to specific conformations through strategic installation of a methyl group. Inhibition studies show that iminosugars with gauche,gauche conformation are 6-10 times more potent than those with gauche,trans conformation, and a manno-configured iminosugar with gauche,gauche conformation is 27 times better inhibitor than 1-deoxymannojirimycin. These results highlight the importance of side chain preorganization for improved inhibitor design in glycosidase inhibition.

Summary: Diabetes and enhanced microbial multidrug resistance due to biofilm formation are major global health problems. Triterpenoids and their derivatives have shown significant contributions in this field. A small library of benzoyl esters of lupeol and beta-amyrin was synthesized and evaluated for their inhibitory potential on pathogenic bacteria biofilms and on alpha-amylase and beta-glucosidase activities. The introduction of benzoyl ester groups at C-3 of lupeol and beta-amyrin substantially improved their antibiofilm and antidiabetic potentials.
