Studying effective column lengths in liquid chromatography of large biomolecules

Summary: Based on the nature of large molecules, only a small segment of a column bed participates in their separation, which is known as on-off elution. Empirical data was investigated to estimate the required length of column bed for effective separation. The study found that for moderately sized solutes (5 - 10 kDa), there is little benefit to using a column bed longer than 3 cm, especially for a gradient of less than 20 minutes. It was also predicted that even shorter columns would be adequate for larger molecules (100 - 150 kDa).

Evaluation of Hold-Up Volume Determination Methods and Markers in Hydrophilic Interaction Liquid Chromatography

Summary: Common methods used in RPLC for hold-up time and volume determination have been tested in HILIC using a zwitterionic ZIC-HILIC column. The pycnometric determination method calculates the overall volume of solvent in the column, including the volume of semi-sorbed eluent on the column packing. The homologous series approach allows accurate determination of hold-up volumes, but is time-consuming. Large neutral markers with poor dipolarity/polarizability and hydrogen bonding interactions can be used as hold-up volume markers in HILIC.

Theoretical and practical comparison of RPLC and RPLC x RPLC: how to consider dilution effects and sensitivity in addition to separation power?

Summary: The objective of this study was to compare one-dimensional reversed-phase liquid chromatography (1D-RPLC) and comprehensive two-dimensional RPLC (RPLC x RPLC) by calculations and experiments. Various quality descriptors were evaluated, and it was found that short columns of 30 x 2.1 mm packed with sub-2-mu m particles should be selected in both dimensions of the 2D-LC setup to obtain the best compromise in terms of peak capacity and sensitivity. The peak capacity in RPLC x RPLC was significantly improved for analysis times beyond 5 min, but the extra-column volume after the second-dimension column was found to be critical for peptides. Higher dilution is not always observed in RPLC x RPLC, and with adequate analytical conditions, RPLC x RPLC can achieve better sensitivity compared to 1D-RPLC.

Volume and composition of semi-adsorb e d stationary phases in hydrophilic interaction liquid chromatography. Comparison of water adsorption in common stationary phases and eluents

Summary: The study used pycnometric and homologous series retention methods to investigate the volume and mean composition of water-rich layers partially adsorbed on the surface of HILIC column fillings with acetonitrile-water and methanol-water as eluents. The findings suggest that water is preferentially adsorbed on the surface of the HILIC bonded phase in hydroorganic eluents with high acetonitrile or methanol concentrations, forming several water-rich transition layers. In eluents with low acetonitrile or methanol concentrations, the amount of preferentially adsorbed water is minimal, and the retention behavior is similar to that of reversed-phase liquid chromatography.

Comparison of existing strategies for keeping symmetrical peaks in on-line Hydrophilic Interaction Liquid Chromatography x Reversed-Phase Liquid Chromatography despite solvent strength mismatch

Summary: This study compared four strategies for method development in on-line HILIC x RPLC, including flow splitting, on-line dilution, ASM dilution, and Total Breakthrough Strategy. The Total Breakthrough Strategy showed the highest peak capacity while on-line dilution using ASM showed the lowest. When separating a tryptic digest in sub-hour separations, peak intensities were generally higher with on-line dilution approaches, but Total Breakthrough Strategy provided approximately 7-fold higher peak intensities compared to flow splitting.
