The northern corn leaf blight resistance gene Ht1 encodes an nucleotide-binding, leucine-rich repeat immune receptor

Shawn Thatcher et al.

Summary: In this study, the causal gene PH4GP-Ht1 underlying resistance to northern corn leaf blight in maize was identified. The PH4GP-Ht1 gene belongs to the NLR gene family and shows a different mode of action compared to previously reported resistance genes. Transgenic plants with PH4GP-Ht1 exhibited a significant reduction in fungal biomass and differential expression of genes associated with defense and photosynthesis. These results provide insights into the molecular basis of resistance to northern corn leaf blight in maize.

The RppC-AvrRppC NLR-effector interaction mediates the resistance to southern corn rust in maize

Ce Deng et al.

Summary: This study reports the molecular cloning and characterization of the SCR resistance gene RppC and the corresponding avirulence effector AvrRppC. Allelic variation of AvrRppC directly determines the effectiveness of RppC-mediated resistance, suggesting that monitoring AvrRppC variants in the field can guide the rational deployment of RppC-containing hybrids in maize production to control SCR.

A pan-genome and chromosome-length reference genome of narrow-leafed lupin (Lupinus angustifolius) reveals genomic diversity and insights into key industry and biological traits

Gagan Garg et al.

Summary: Narrow-leafed lupin is a high-protein grain suitable for sustainable farming. This study provides a reference genome and pan-genome assembly for narrow-leafed lupin, compares it with the genome of its close relative white lupin, and identifies key alkaloid regulatory genes and the deficiency of disease resistance genes in the narrow-leafed lupin genome. These findings offer new opportunities for lupin crop improvement.

Cloning southern corn rust resistant gene RppK and its cognate gene AvrRppK from Puccinia polysora

Gengshen Chen et al.

Summary: This study reports that a maize gene RppK confers resistance to southern corn rust and can recognize the effector AvrRppK produced by Puccinia polysora. The introgression of RppK into maize enhances resistance against multiple isolates of P. polysora, thereby increasing yield in the presence of southern corn rust.

A super pan-genomic landscape of rice

Lianguang Shang et al.

Summary: By studying the pan-genomes of large natural populations, we can uncover the genetic diversity and complexity of rice. We have created a super pan-genome of rice using long-read assembly and identified extensive structural variations and gene presence/absence variations. Additionally, we have discovered genetic variants associated with various traits in rice and identified a common set of genes that have played a role in adaptation and domestication in Asian and African rice.

Genome-wide functional analysis of hot pepper immune receptors reveals an autonomous NLR clade in seed plants

Hye-Young Lee et al.

Summary: This study identified a group of NLRs (ANLs) carrying autoactive coiled-coil domains in pepper plants, which may function as singleton NLRs. The research also revealed that autoactive coiled-coil domains are well conserved in Solanaceae plants and rice, a monocot plant. Furthermore, phylogenetic analyses showed that ANLs are present in all tested seed plants.

A designer rice NLR immune receptor confers resistance to the rice blast fungus carrying noncorresponding avirulence effectors

Yang Liu et al.

Summary: Plant NLR receptors can recognize avirulence effectors either directly through integrated domains or indirectly via effector-targeted proteins. Designer NLR receptors have been successfully generated by engineering integrated domains or targeted proteins based on previous knowledge. This study demonstrates that engineered rice NLR receptor carrying an integrated HMA domain can confer resistance to noncorresponding but structurally similar MAX effectors with unknown mechanisms, providing a practical approach for developing resistant cultivars.

Alleles of a wall-associated kinase gene account for three of the major northern corn leaf blight resistance loci in maize

Ping Yang et al.

Summary: The study reveals that Ht2 and Ht3 are identical and allelic to the previously cloned Htn1 gene, all of which are allelic to the wall-associated receptor-like kinase gene ZmWAK-RLK1. These three genes exhibit polymorphisms in different maize lines, affecting resistance responses. The classic description of these genes may have been confounded by genetic background effects in the past.

Summary: In this study, a genome-wide analysis of NLR genes in barley was conducted, revealing a large number of NLR genes, including RNL and CNL subtypes. The majority of NLR genes were found in multigene clusters, and phylogenetic analysis identified at least 18 ancestral CNL lineages shared between barley, T. urartu, and Arabidopsis thaliana.

Analysis of intraspecies diversity reveals a subset of highly variable plant immune receptors and predicts their binding sites

Daniil M. Prigozhin et al.

Summary: The study investigated the NLR complements of Arabidopsis thaliana and Brachypodium distachyon, identifying a limited number of NLR subfamilies with high allelic diversity. The analysis revealed that predicted specificity-determining residues cluster on the surfaces of Leucine-Rich Repeat domains, but the locations of the clusters vary among NLR subfamilies. By comparing NLR phylogeny, allelic diversity, and known functions, a hypothesis for the emergence of direct and indirect pathogen-sensing receptors and autoimmune NLRs was formulated.

Summary: Despite the prevalence and recurrence of polyploidization in flowering plants, its impacts on crop intraspecific genome diversification remain largely unknown. This study investigates the consequences of whole-genome triplication in Brassica rapa and uncovers the potential contributions of allopolyploidization on intraspecific diversification. Additionally, the role of structural variations in favorable trait domestication is explored.
