An atlas of substrate specificities for the human serine/threonine kinome

Jared L. Johnson et al.

Summary: Protein phosphorylation is a common post-translational modification, and thousands of phosphorylated sites have been associated with human diseases and biological processes. However, the responsible protein kinases for most phosphorylation events are currently unknown. This study used synthetic peptide libraries to profile the substrate specificity of 303 Ser/Thr kinases and identified the kinases capable of phosphorylating reported phosphorylation sites.

Loss of LKB1-NUAK1 signalling enhances NF-κB activity in a spheroid model of high-grade serous ovarian cancer

Adrian Buensuceso et al.

Summary: In this study, the researchers found that loss of Liver kinase B1 (LKB1) and its substrate NUAK1 in high-grade serous ovarian cancer (HGSOC) leads to hyperactive NF-kappa B signaling, which suppresses reactive oxygen species (ROS) production and supports cell survival. These findings suggest that combined inhibition of NUAK1 and NF-kappa B signaling may be a novel therapeutic strategy for advanced HGSOC.

NUAK1 promotes organ fibrosis via YAP and TGF-β/SMAD signaling

Tianzhou Zhang et al.

Summary: Fibrosis is a key driver of multiple chronic diseases, but effective therapies are lacking. The study identifies NUAK1 as a profibrotic kinase that is up-regulated in fibrotic organs. NUAK1 promotes fibrosis through positive feedback loops involving YAP and TGF-β/SMAD signaling. Inhibiting NUAK1 can attenuate fibrosis and represents a potential therapeutic target.

Discovery of a novel NUAK1 inhibitor against pancreatic cancer

Myeong-Seong Seo et al.

Summary: The novel NUAK1 inhibitor, KI-301670, effectively inhibits pancreatic cancer growth and proliferation, induces cell cycle arrest and apoptotic effect, and inhibits migration and invasion of cancer cells by targeting the PI3K/AKT pathway. It also shows significant tumor growth inhibition in a mouse xenograft tumor model, suggesting its potential as a therapeutic candidate for pancreatic cancer treatment.

Development and Therapeutic Potential of NUAKs Inhibitors

Muhammad Faisal et al.

Summary: NUAK isoforms, NUAK1 (ARK5) and NUAK2 (SNARK), are crucial members of the AMPK family of protein kinases, associated with various pathological disorders. Targeting NUAK isoforms with inhibitors shows promise in treating metabolic diseases and cancers, with a focus on their involvement in key biological processes and disease progression. Numerous small molecule inhibitors for NUAK isoforms have been identified through critical literature review and analysis.

Heterogeneity in Pancreatic Cancer Fibroblasts-TGFβ as a Master Regulator?

Dale M. Watt et al.

Summary: Pancreatic ductal adenocarcinoma is an aggressive disease with limited available therapies, notable for its high tumour complexity and recent findings on the intricacy of signalling within the stroma, particularly the molecular and functional heterogeneity of cancer associated fibroblasts. While some progress has been made, there are still considerable gaps in our knowledge that require further research efforts.
