Regulation and function of the mammalian tricarboxylic acid cycle

Paige K. Arnold et al.

Summary: The tricarboxylic acid (TCA) cycle, also known as the Krebs cycle, is a central metabolic pathway that oxidizes nutrients to support cellular energy production. Recent studies have shown that the TCA cycle is dynamic and can be utilized in cancer and other pathological states. This review provides a comprehensive overview of the TCA cycle, including its origins, discovery, requirements for sustained function, and regulatory nodes. It also discusses recent advances in understanding the flexibility and heterogeneity of TCA cycle activity in mammalian cells, emphasizing the importance of differential regulation and configuration to meet the specific requirements of distinct cell states.

Multiomics analyses reveal early metabolic imbalance and mitochondrial stress in neonatal photoreceptors leading to cell death in Pde6b(rd1/rd1) mouse model of retinal degeneration

Ke Jiang et al.

Summary: Retinal diseases are genetically heterogeneous and have complex etiology. Mutations in over 200 genes can lead to dysfunction and death of photoreceptor cells in retinal neurodegeneration. A study on a mouse model of retinitis pigmentosa found that mitochondrial damage and metabolic disruptions are early drivers of photoreceptor cell death in retinal degeneration.

Retinal Layer Separation (ReLayS) method enables the molecular analysis of photoreceptor segments and cell bodies, as well as the inner retina

Vyara Todorova et al.

Summary: Understanding the physiology of the retina, particularly the highly polarized photoreceptors, is crucial for advancing our knowledge of normal vision processes and developing effective therapies for retinal degenerative diseases. However, studying the molecular mechanisms of photoreceptors is challenging due to the heterogeneity of retinal tissue and the lack of reliable cell separation methods. In this study, the researchers present a simple technique called ReLayS that successfully separates different parts of photoreceptors, enabling protein and RNA analysis. The method was validated using known proteins and genes and also revealed light-driven translocation of transducin.

Structural comparison of GLUT1 to GLUT3 reveal transport regulation mechanism in sugar porter family

Tania Filipa Custodio et al.

Summary: The study compared the structures and functional data of GLUT1 and GLUT3, revealing significant differences in glucose affinity between them, possibly due to interactions between the cytosolic SP motif and conserved A motif. Additionally, previously undescribed Cl- ion site and endofacial lipid/glucose binding site in GLUT1 were identified as modulators of GLUT kinetics.

Inhibition of the HIF-1α/BNIP3 pathway has a retinal neuroprotective effect

Hiromitsu Kunimi et al.

Summary: In this study, using retina-specific Hif-1 alpha conditional knockout (cKO) mice revealed that the reduction of Hif-1 alpha can lead to a decrease in inner retinal neurodegeneration. Experiments showed that depletion of Bnip3 in retinal neurons can have neuroprotective effects, supporting the critical role of the HIF-1 alpha/BNIP3 pathway in inner retinal neurodegeneration.

Transcriptomic analysis of the mouse retina after acute and chronic normobaric and hypobaric hypoxia

L. J. A. Ebner et al.

Summary: This study investigates the impact of chronic oxygen deficiency on the retina, revealing that retinal cells have the ability to adapt to long-term hypoxia and that the consequences of hypobaric hypoxia differ from those of normobaric hypoxia. The findings provide insights into the molecular changes in response to various hypoxic conditions and can serve as references for validating and comparing retinal disease models associated with hypoxia.

Replenishment of TCA cycle intermediates provides photoreceptor resilience against neurodegeneration during progression of retinitis pigmentosa

Ashley A. Rowe et al.

Summary: The study found that reducing TCA cycle intermediates in RP disease led to cell degeneration, but promoting citrate production protected photoreceptor cells and prolonged visual function. This therapeutic effect was not specific to certain genetic mutations, showing broad applicability for RP and other retinal degenerative diseases. Targeting TCA cycle activity in the neural retina promoted photoreceptor survival and visual function during neurodegenerative disease.
