The S1PR2-CCL2-BDNF-TrkB pathway mediates neuroinflammation and motor incoordination in hyperammonaemia

Yaiza M. Arenas et al.

Summary: Chronic hyperammonaemia induces neuroinflammation and motor incoordination in hepatic encephalopathy, and this study identifies the involvement of the S1PR2-CCL2-BDNF-TrkB pathway. Blocking S1PR2 can alleviate neuroinflammation and restore motor coordination.

Celastrol exerts a neuroprotective effect by directly binding to HMGB1 protein in cerebral ischemia-reperfusion

Dan-Dan Liu et al.

Summary: In this study, Cel was found to inhibit the proinflammatory activity of HMGB1 by directly binding to it, blocking the binding of HMGB1 to its inflammatory receptors in the ischemic and hypoxic microenvironment. Additionally, Cel exhibited neuroprotective and anti-inflammatory effects by targeting HSP70 and NF-kappa B p65, leading to neuronal rescue from OGD injury in vitro and decreased cerebral infarction in vivo.

Fecal Microbiota Transplantation Exerts a Protective Role in MPTP-Induced Parkinson's Disease via the TLR4/PI3K/AKT/NF-κB Pathway Stimulated by α-Synuclein

Zhe Zhong et al.

Summary: The study showed that fecal microbiota transplantation (FMT) can alleviate motor impairment, reduce fecal SCFAs levels, decrease alpha-syn expression, and inhibit microglial activation in the substantia nigra (SN) of PD mice. FMT also blocked the TLR4/PI3K/AKT/NF-kappa B signaling pathway in the SN and striatum, providing protection against PD.

Long noncoding RNA BS-DRL1 modulates the DNA damage response and genome stability by interacting with HMGB1 in neurons

Min-Min Lou et al.

Summary: This study demonstrates a neuron-specific role for Brain Specific DNA-damage Related lncRNA1 (BS-DRL1) in modulating DNA damage response (DDR) by interacting with HMGB1 in a cell-type specific manner. Knockout mice of BS-DRL1 show impaired motor function and purkinje cell degeneration, indicating a potential protective role of lncRNA against neurodegeneration.

Tau oligomer induced HMGB1 release contributes to cellular senescence and neuropathology linked to Alzheimer's disease and frontotemporal dementia

Sagar Gaikwad et al.

Summary: The study demonstrates that TauO-induced neuroinflammation is primarily mediated through HMGB1 release and inflammatory SASP, leading to astrocyte senescence. Inhibition of HMGB1 release can prevent TauO-induced cellular senescence, reduce neuroinflammation, and improve cognitive function.
