Cognitive function after concurrent temozolomide-based chemoradiation therapy in low-grade gliomas

Deborah Y. Park et al.

Summary: This study evaluated the effects of concurrent temozolomide-based chemoradiation therapy on neurocognitive function in low-grade glioma patients. Patients aged over 40 and with education over 16 years demonstrated superior baseline verbal memory. Overall, neurocognitive function remained stable following the treatment.

KDM5B promotes tumorigenesis of Ewing sarcoma via FBXW7/CCNE1 axis

Binbin Chen et al.

Summary: Upregulated histone demethylase KDM5B in Ewing sarcoma (EwS) was found to be associated with poor prognosis. KDM5B is involved in regulating the cell cycle pathway and affects the accumulation of CCNE1 protein. Targeting KDM5B with epigenetic drugs could be a potential treatment for EwS.

The BASP1 transcriptional corepressor modifies chromatin through lipid-dependent and lipid-independent mechanisms

Alexander J. Moorhouse et al.

Summary: The transcriptional corepressor BASP1 requires N-terminal myristoylation for its activity and functions through interactions with nuclear lipids. Removal of active histone modifications H3K9ac and H3K4me3 by BASP1 requires N-terminal myristoylation, while placement of the repressive histone modification H3K27me3 does not require lipidation. BASP1 regulates activity of multiple transcription factors and induces extensive changes in chromatin accessibility, with about 50% of BASP1 target genes showing lipidation-dependent chromatin compaction and transcriptional repression.

Intrinsic immune evasion patterns predict temozolomide sensitivity and immunotherapy response in lower-grade gliomas

Zewei Tu et al.

Summary: Intrinsic immune-evasion patterns of gliomas can help predict clinical prognosis and determine treatment. Gene expression analysis identified three subgroups of lower-grade glioma (LGG) with distinct prognosis and immune infiltrations. Assessing immune-evasion levels can guide individualized therapeutic strategies for LGG patients.

Argininosuccinate lyase drives activation of mutant TERT promoter in glioblastomas

Zhumei Shi et al.

Summary: This study reveals a previously unrecognized mechanistic insight into the epigenetic activation of mutant TERT promoters, where ASL, which interacts with GABPA, plays a crucial role. EGFR activation induces ASL phosphorylation, leading to enhanced TERT expression through the interaction between ASL and GABPA. The generated fumarate from ASL inhibits KDM5C, resulting in increased trimethylation of histone H3 Lys4, which facilitates the recruitment of c-Myc to TERT promoters.

Current status and recent advances in reirradiation of glioblastoma

Giuseppe Minniti et al.

Summary: Reirradiation has shown survival benefits for patients with recurrent GBM, but questions remain about the efficacy and toxicity of a second course of radiation. Current clinical discussions on reirradiation of GBM focus on appropriate patient selection, radiation technique, optimal dose fractionation, brain tolerance to reirradiation, and the risk of radiation necrosis.

High BASP1 Expression is Associated with Poor Prognosis and Promotes Tumor Progression in Human Lung Adenocarcinoma

Xin Wang et al.

Summary: BASP1 plays a tumor-promoting role in lung adenocarcinoma by inhibiting cell apoptosis and activating the Wnt/beta-catenin pathway, leading to increased proliferation and migration of cancer cells. This overexpression is associated with poor prognosis in lung adenocarcinoma.

Immunotherapy for recurrent glioblastoma: practical insights and challenging prospects

Xin Wang et al.

Summary: For recurrent glioblastoma (rGB) patients, neoadjuvant anti-PD-1 therapy followed by surgery and adjuvant anti-PD-1 therapy can significantly improve overall survival. Additionally, tumor-specific peptide vaccination or CAR-T cell therapy has shown promise for treating rGB patients. Effective predictive biomarkers for clinical efficacy are needed, along with efforts to enhance T cell function through engineering techniques.

Cholesterol is required for transcriptional repression by BASP1

Amy E. Loats et al.

PARP-mediated PARylation of MGMT is critical to promote repair of temozolomide-induced O6-methylguanine DNA damage in glioblastoma

Shaofang Wu et al.

Summary: This study demonstrates the critical role of PARylation of MGMT by PARP in repairing TMZ-induced O-6-MetG, with PARP inhibitors reducing MGMT function and sensitizing GBM to TMZ. The findings provide a rationale for combining PARP inhibitors to enhance TMZ sensitivity in MGMT-unmethylated GBM.
