Expanding the molecular spectrum of gene fusions in endometrial stromal sarcoma: Novel subunits of the chromatin remodeling complexes PRC2 and NuA4/TIP60 as alternative fusion partners

Josephine K. K. Dermawan et al.

Summary: In this study, we report a series of endometrial stromal sarcoma (ESS) cases with novel gene fusions. These cases demonstrate morphological and molecular heterogeneity. The identified fusion genes play important roles in the regulation of the PRC2 and/or NuA4/TIP60 complexes.

Recurrent KAT6B/A::KANSL1 Fusions Characterize a Potentially Aggressive Uterine Sarcoma Morphologically Overlapping With Low-grade Endometrial Stromal Sarcoma

Abbas Agaimy et al.

Summary: With the widespread use of next-generation sequencing, the genetic landscape of uterine mesenchymal neoplasms is rapidly evolving. This study identified 13 uterine stromal neoplasms carrying a KAT6B:: KANSL1 fusion, highlighting the importance of understanding the molecular subtypes of these tumors.

NTRK Fusions in a Sarcomas Series: Pathology, Molecular and Clinical Aspects

Vasiliki Siozopoulou et al.

Summary: Targeted therapy against tyrosine receptor kinase has shown promising response rates and durable effects in tumors with specific gene fusions. However, current diagnostic methods for these tumors are limited. In this study, the immunohistochemical expression of pan-TRK was investigated in soft tissue and bone sarcomas, leading to the identification of some cases with NTRK gene fusions. Further analysis revealed unique histological, molecular, and clinical findings in these fused sarcomas. The study aims to expand knowledge of NTRK-fused sarcomas and correlate their characteristics with clinical outcomes.
