Lithium therapy subdues neuroinflammation to maintain pyramidal cells arborization and rescues neurobehavioural impairments in ovariectomized rats

Anil Kumar Rana et al.

Summary: Estrogen deprivation during menopause leads to changes in brain neuronal circuit and neurobehavioral symptoms later in life. Hormone replacement therapy can partially alleviate these abnormalities but is associated with various adverse effects. This study investigated the effect of lithium therapy on neurobehavioral impairments in a rat model of ovariectomy, which mimics postmenopausal conditions. The results showed that lithium treatment protected against spatial and recognition memory impairments, as well as depression-like behavior in ovariectomized rats. Additionally, lithium treatment increased dendritic length and spine density in certain brain regions, reduced proinflammatory markers, normalized reactive astrogliosis, and modulated key signaling pathways. These findings suggest that lithium may be a potential therapeutic agent for managing postmenopausal neurological symptoms.

Reactive astrocytes acquire neuroprotective as well as deleterious signatures in response to Tau and Aβ pathology

Zoeb Jiwaji et al.

Summary: Alzheimer's disease affects astrocytes, and both A beta and Tau pathology induce changes in astrocytes. A beta mainly influences the expression of AD risk genes, while both pathologies induce age-dependent changes and have overlapping features with astrocytes found in post-mortem AD patients. Both A beta and Tau pathology alter the characteristics of astrocytes, including repression of bioenergetic and translation machinery, induction of inflammation pathways, and expression of protein degradation/proteostasis genes. Astrocyte-specific expression of Nrf2, a protective factor, can recapitulate elements of these astrocyte changes and reduce A beta deposition and phospho-tau accumulation. This Nrf2 expression also improves brain-wide transcriptional deregulation, cellular pathology, neurodegeneration, and behavioral/cognitive deficits.

Deciphering key regulators involved in epilepsy-induced cardiac damage through whole transcriptome and proteome analysis in a rat model

Supriya Sharma et al.

Summary: This study identified STAT3, Mapk8, and Erbb as key regulators involved in seizure-associated cardiac changes using an integrated approach of RNA-Seq, proteomics, and system biology analysis in a rat model of temporal lobe epilepsy. The study also supported the activation of the transforming growth factor beta (TGF-beta) pathway in the heart of epileptic animals. Gene and protein expression studies further established a key role of STAT3, Erbb, and Mapk8 in the development of cardiac changes linked with recurrent seizures.

Docosahexanoic acid signals through the Nrf2-Nqo1 pathway to maintain redox balance and promote neurite outgrowth

Jennifer Drolet et al.

Summary: Evidence suggests that n-3 polyunsaturated fatty acids can activate the Nrf2 antioxidant pathway, promoting neuronal differentiation and protecting against agrochemical-induced neuritic retraction. DHA-mediated activation of Nrf2 leads to neurite outgrowth and protection, mainly through induction of the Nrf2-target gene NQO1.

Risks, Benefits, and Treatment Modalities of Menopausal Hormone Therapy: Current Concepts

Jaya Mehta et al.

Summary: Prescribing practices for menopausal hormone therapy have evolved in recent decades based on changing understanding of the treatment's risks and benefits. Recent research suggests that the risks of HT are low for healthy women under 60 or within ten years of menopause, while the benefits may outweigh the risks for those experiencing bothersome vasomotor symptoms. HT also plays a role in preventing osteoporosis for appropriate candidates.
