A Robust ROS Generation Strategy for Enhanced Chemodynamic/Photodynamic Therapy via H2O2/O2 Self-Supply and Ca2+ Overloading

The CaZFCP releases CaO2 nanoparticles, Fe2+, and photosensitizer Chlorin e6 (Ce6) in the weakly acidic tumor microenvironment, alleviating the deficiency of endogenous H2O2 and hypoxia in the tumor area, leading to enhanced production of hydroxyl radicals and singlet oxygen for improved CDT and PDT. Additionally, the Ca2+ overloading induced by CaO2 decomposition amplifies intracellular oxidative stress, resulting in mitochondrial dysfunction and further enhancing the efficacy of combined CDT/PDT. The in vitro and in vivo experimental results confirm the excellent tumor inhibition effect of CaZFCP, providing a simple paradigm for ROS-involved cancer therapies.
