Summary: This study investigated the effects of a model DIO inhibitor on a laboratory model species, Xenopus laevis, to understand the linkages between in vitro potency, in vivo biochemical responses, and adverse organismal outcomes. The results showed that the inhibitor could inhibit DIO in vitro and cause delayed metamorphosis and reduced growth in the exposed amphibians.

Summary: The iodide recycling enzyme, iodotyrosine deiodinase (IYD), is an understudied molecular mechanism through which environmental chemicals can potentially cause thyroid disruption. A screening assay was developed and used to identify 194 IYD inhibitors, with 154 chemicals further tested for potency. This study expands the scope of thyroid-relevant molecular targets for chemical screening and aids in prioritizing chemicals for targeted in vivo testing for thyroid-related adverse outcomes.

Summary: Deiodinase enzymes play a critical role in controlling activation or inactivation of thyroid hormones during vertebrate development. Screening of chemicals against human and Xenopus laevis deiodinase enzymes revealed potential specific inhibitors, with human assay results showing predictive value for amphibian activity. This study paves the way for future research on cross-species sensitivity to chemicals using recombinant non-mammalian proteins.
