MBG/PGA-PCL composite scaffolds provide highly tunable degradation and osteogenic features

Summary: Achieving a balance between biodegradability and osteogenic capacity in biosynthetic bone grafts remains a challenge. In this study, we addressed this challenge by incorporating mesoporous bioactive glass (MBG) into poly(caprolactone-co-glycolide) (PGA-PCL) at gradient ratios. The MBG/PGA-PCL (PGC/M) scaffolds exhibited a highly porous structure with incorporated MBGs uniformly dispersed. The incorporation of MBG significantly improved the scaffold's surface properties, apatite-formation ability, pH stability, and promoted the attachment, spreading, viability, and proliferation of bone marrow stromal cells (BMSCs) on the scaffolds. Moreover, the scaffolds demonstrated enhanced ALP activity and osteogenesis-related gene expression in a dose-dependent manner. When implanted into rat cranial bone defects, the scaffolds with higher MBG incorporation ratios induced significantly better bone repair. These findings suggest that the PGC/M scaffolds are promising biomaterials for bone repair with tunable properties.

Proportion-dependent osteogenic activity of electrospun nano-hydroxyapatite/polylactic acid fiber membrane in vitro and in vivo

Summary: The study investigated the effect of proportion on the physicochemical and osteogenic properties of electrospun nano-hydroxyapatite/polylactic acid (nHAP/PLA) fiber membrane. The addition of an appropriate amount of nHAP increased the hydrophilicity of the fiber surface and enhanced the modulus in the perpendicular direction to the membrane plane. In biological experiments, 6-10 wt% nHAP/PLA membrane significantly promoted cell proliferation and differentiation. In vivo experiments showed that the addition of 6-10 wt% nHAP/PLA promoted cell recruitment and infiltration, and increased bone tissue volume/total tissue volume and bone mineral density.
