Hyperpolarized pyruvate to measure the influence of PKM2 activation on glucose metabolism in the healthy kidney

Lotte Bonde Bertelsen et al.

Summary: The PKM2 activator TEPP-46 increases glycolytic activity in the kidneys, leading to increased lactate production as seen by hyperpolarized pyruvate-to-lactate conversion. These metabolic changes, largely independent of renal function, could represent a highly sensitive metabolic readout for future drugs targeting PKM2. Further studies are warranted in disease models to evaluate if [1-C-13]pyruvate-to-[1-C-13]lactate conversion can predict treatment outcome.
