A computational method for predicting the aggregation propensity of IgG1 and IgG4(P) mAbs in common storage buffers

James T. T. Heads et al.

Summary: The propensity for some monoclonal antibodies to aggregate at physiological and manufacturing pH values can hinder their use as therapeutic molecules or delay their market introduction. In this study, the researchers developed a computational method, called MAPT, to accurately predict the aggregation propensity of antibodies in common storage buffers. The method combines isotype-dependent, charge-based models with a hydrophobicity descriptor derived from a homology model, resulting in a robust indicator for antibody developability.

High-throughput profiling of antibody self-association in multiple formulation conditions by PEG stabilized self-interaction nanoparticle spectroscopy

Samantha Phan et al.

Summary: Affinity-capture self-interaction nanoparticle spectroscopy (AC-SINS) is a test method developed for evaluating the colloidal stability of antibodies, commonly used for screening antibodies in the early stages of pharmaceutical discovery. We have made improvements to the method by using a stabilizing reagent to broaden the range of formulation conditions that can be screened, and by introducing a new algorithm and data processing schema for streamlined data analysis. The optimized test format expands the screening applicability to critical formulation conditions and is supported by a custom data management workflow for efficient data extraction, analysis, and automation.

Functional Changes of Therapeutic Antibodies upon Exposure to Pro-Oxidative Agents

Maxime Lecerf et al.

Summary: Therapeutic monoclonal antibodies have had a transformative impact on clinical practice, but developing therapeutic antibodies remains complex. This study revealed the vulnerability of immunoglobulins to oxidation and the development of polyreactivity in a substantial number of therapeutic antibodies. The induction of polyreactivity was found to be correlated with somatic mutations in the variable domains of immunoglobulin chains. These findings can contribute to the selection of antibody therapeutics with suitable developability profiles.
