An autopsy case of progressive supranuclear palsy treated with monoclonal antibody against tau

Deep learning-based model for diagnosing Alzheimer's disease and tauopathies

Summary: This study developed a deep learning model for differentiating tauopathies on tau-immunostained digital slide images. Using CP13-immunostained slide images for training, a random forest classifier achieved an average test score of 0.97, correctly diagnosing 29 out of 30 cases. Validation with hold-out datasets showed over 92% diagnostic accuracy without data augmentation and over 95% with augmentation.

CSF tau microtubule-binding region identifies pathological changes in primary tauopathies

Summary: Currently, there are no fluid biomarkers or imaging tracers available for the diagnosis and treatment of other tauopathies. This study identifies specific tau species that increase in the brains of patients with CBD, PSP, FTLD-MAPT, and AD, but decrease in their cerebrospinal fluid. Measurements of these specific tau species in cerebrospinal fluid can differentiate between different diseases and aid in the diagnosis and clinical trial design of primary tauopathies.

It is associated with various underlying pathologies such as Corticobasal degeneration (CBD), progressive supranuclear palsy (PSP), Alzheimer's disease (AD), and frontotemporal lobar degeneration with TDP-43 inclusions. Regardless of the underlying pathology, CBS is associated with frontoparietal, motor and premotor pathology.

Unleashing the potential of digital pathology data by training computer-aided diagnosis models without human annotations

Summary: The study proposes an approach to train computer-aided diagnosis tools in digital pathology without the need for manual annotations, by automatically extracting meaningful concepts from diagnostic reports as weak labels to train convolutional neural networks for histopathology diagnosis. The approach is trained on 3,769 clinical images and reports from two hospitals and tested on over 11,000 images from private and publicly available datasets, showing that text analysis combined with end-to-end deep neural networks can achieve solid performance without manual annotations.

Interpretable deep learning of myelin histopathology in age-related cognitive impairment

Summary: Age-related cognitive impairment is a complex condition with multiple pathological factors. In this study, a weakly supervised multiple instance learning algorithm was used to predict cognitive impairment based on whole slide images of brain tissue sections. Attention analysis helped identify the underlying subregional architecture and cellular features associated with cognitive impairment. The results demonstrate the potential of interpretable deep learning in identifying unexpected aspects of pathology in cognitive impairment and other neurobiological disorders.
