Understanding and managing uncertainty and variability for wastewater monitoring beyond the pandemic: Lessons learned from the United Kingdom national COVID-19 surveillance programmes

Matthew J. Wade et al.

Summary: The COVID-19 pandemic has led to unprecedented pressure on global public health resources, prompting the development of National wastewater surveillance programmes in the UK to monitor the SARS-CoV-2 virus. This work highlights the unique importance of wastewater-based epidemiology (WBE) for public health protection, with potential value beyond COVID-19 for monitoring a range of health markers.

A comparison of precipitation and filtration-based SARS-CoV-2 recovery methods and the influence of temperature, turbidity, and surfactant load in urban wastewater

Jessica L. Kevill et al.

Summary: Wastewater-based epidemiology (WBE) is a useful tool in monitoring SARS-CoV-2 during the pandemic. This study compared three viral concentration methods and found that there was no major difference in viral recovery using these methods. However, factors such as sample turbidity, storage temperature, and surfactant load did affect viral recovery, emphasizing the importance of considering these factors when working with wastewater samples.

Monitoring occurrence of SARS-CoV-2 in school populations: A wastewater-based approach

Victor Castro-Gutierrez et al.

Summary: This study used wastewater-based epidemiology to monitor the presence of the COVID-19 virus in 16 schools in England. Approximately 47.3% of the analyzed wastewater samples tested positive for the virus genes, demonstrating the potential of wastewater-based epidemiology as a supplementary tool for clinical testing.

Accounting for Dilution of SARS-CoV-2 in Wastewater Samples Using Physico-Chemical Markers

Henry Wilde et al.

Summary: This paper presents a novel method for obtaining real-time estimates of wastewater dilution and total daily volume through wastewater treatment works. The method, which uses several physico-chemical markers in the wastewater, proves effective in recovering capped peaks in wastewater flow measurements and normalizing SARS-CoV-2 viral loads in wastewater samples.

Detection of SARS-CoV-2 in raw and treated wastewater in Germany - Suitability for COVID-19 surveillance and potential transmission risks

Sandra Westhaus et al.

Summary: Wastewater-based monitoring showed the presence of SARS-CoV-2 genetic traces in different raw wastewaters in North Rhine-Westphalia, Germany. However, it was suggested that wastewater might not be a major route for transmission to humans. Further validation for enveloped viruses in wastewater is needed.

SARS-CoV-2 has been circulating in northern Italy since December 2019: Evidence from environmental monitoring

Giuseppina La Rosa et al.

Summary: Italy had SARS-CoV-2 circulating as early as the end of 2019, with the virus spreading in different geographic regions simultaneously. The study emphasizes the importance of environmental surveillance as an early warning system to monitor virus levels and identify outbreaks before they are officially reported to the healthcare system.

Application of multi-parameter population model based on endogenous population biomarkers and flow volume in wastewater epidemiology

Chenzhi Hou et al.

Summary: In this study, the consumption or prevalence of acesulfame, caffeine, paracetamol and amantadine in Hebei province, China was estimated using wastewater-based epidemiology. A multi-parameter population model was established to improve population estimation accuracy, showing high correlations and low variation compared to census data. The estimated results were consistent with previous research and sales data calculations, demonstrating the feasibility and potential for lower uncertainties in population estimation using a multi-parameter population model.

Uncertainties in estimating SARS-CoV-2 prevalence by wastewater-based epidemiology

Xuan Li et al.

Summary: Wastewater-based epidemiology (WBE) is a promising approach for estimating COVID-19 prevalence by detecting SARS-CoV-2 RNA in wastewater, but methodological challenges could impact accuracy. Dividing WBE into five steps, this study quantified uncertainties at each step and found that using high-frequency flow-proportional sampling and actual water usage data reduced overall uncertainties to 20-40%, with analytical uncertainty being the dominant factor. Using surrogate viruses and improving analytical approaches could enhance reliability of WBE for monitoring COVID-19 prevalence.

The presence of SARS-CoV-2 RNA in human sewage in Santa Catarina, Brazil, November 2019

Gislaine Fongaro et al.

Summary: SARS-CoV-2 RNA was detected in human sewage samples from Florianopolis, Brazil from November 2019 to March 2020, earlier than the first reported case in the Americas. This suggests that the virus may have been circulating undetected in the community.

On forecasting the community-level COVID-19 cases from the concentration of SARS-CoV-2 in wastewater

Yongtao Cao et al.

Summary: This study aimed to investigate the association between SARS-CoV-2 wastewater concentrations and COVID-19 cases at the community-level, and to assess the integration of this data into a wastewater-based epidemiological statistical model for accurate forecasting of COVID-19 infections. The results show that a VAR(1) model estimated with the original data has the strongest forecasting ability, with forecast accuracy for 1 week, 2 weeks, and 3 weeks in the future ranging from 8.97% to 21.57%.

A multicenter study investigating SARS-CoV-2 in tertiary-care hospital wastewater. viral burden correlates with increasing hospitalized cases as well as hospital-associated transmissions and outbreaks

Nicole Acosta et al.

Summary: This study utilized wastewater-based detection of SARS-CoV-2 to assess the epidemiology of SARS-CoV-2 in hospitals. The results showed an increasing trend of N1 and N2 genes in wastewater over time in hospitals with single monitoring points, correlating with the rise in hospitalizations. Despite increasing hospitalizations, the detection of SARS-CoV-2 N1-RNA in wastewater was able to differentiate nosocomial-acquired cases of COVID-19 and unit-specific outbreaks. Wastewater-based monitoring of SARS-CoV-2 is a promising tool for passive surveillance and case identification, containment, and mitigation in acute-care medical facilities.
