TAK1 accelerates transplant arteriosclerosis in rat aortic allografts by inducing autophagy in vascular smooth muscle cells

Xichuan Zheng et al.

Summary: TAK1 signaling pathway plays a crucial role in the proliferation and migration of vascular smooth muscle cells (VSMCs). This study reveals that activation of TAK1 controls autophagy activation and regulates cell cycle, proliferation, and migration responses in VSMCs. This provides a potential molecular mechanism and therapeutic target for preventing transplant vasculopathy.

Autophagy Is Differentially Regulated in Leukocyte and Nonleukocyte Foam Cells During Atherosclerosis

Sabrina Robichaud et al.

Summary: This study compared autophagy flux in lipid-rich aortic intimal populations to determine whether VSMC-derived foam cells metabolize LDs similarly to their macrophage counterparts. The findings indicate that VSMC and macrophage foam cells have distinct mechanisms of cholesterol efflux, with impaired autophagy flux in VSMC foam cells that can be induced by pharmacological means. Further research targeting autophagy specifically in VSMC foam cells is warranted to promote the resolution of atherosclerotic plaques.

Non-canonical WNT signalling in cardiovascular disease: mechanisms and therapeutic implications

Ioannis Akoumianakis et al.

Summary: Non-canonical WNT signaling plays an important role in cardiovascular diseases such as atherosclerosis, heart failure, and arrhythmias. This review provides an overview of non-canonical WNT signaling, describes its links to the pathogenesis of these cardiovascular diseases, and explores the clinical potential of targeting individual components of non-canonical WNT signaling for their treatment.
