MAPK1/3 kinase-dependent ULK1 degradation attenuates mitophagy and promotes breast cancer bone metastasis

Rong Deng et al.

Summary: ULK1 deficiency impairs mitophagy in breast cancer cells under hypoxia, leading to accumulation of damaged mitochondria, NLRP3 inflammasome activation, and bone metastasis. Phosphorylation of ULK1 triggers its degradation, and a negative correlation between ULK1 expression levels and p-MAPK1/3 was observed. The use of MAP2K/MEK inhibitor trametinib can restore mitophagy function and inhibit NLRP3 inflammasome activation to reduce bone metastasis.

EpCAM-Mediated Cellular Plasticity Promotes Radiation Resistance and Metastasis in Breast Cancer

Arijit Mal et al.

Summary: A substantial number of breast cancer patients undergoing radiation therapy develop local recurrence over time, suggesting adaptive radioresistance. This study established radioresistant cell lines and found that EpCAM overexpression in CSCs contributes to radioresistance and metastatic behavior. EpCAM-overexpressing cells exhibit plasticity and heterogeneity, ultimately aggravating the disease condition by inducing radioresistant and metastatic behavior.

Filopodium-derived vesicles produced by MIM enhance the migration of recipient cells

Tamako Nishimura et al.

Summary: Research has shown that I-BAR proteins, especially MIM, generate large EVs by scission of filopodia, containing lysophospholipids and proteins that stimulate cell migration. These filopodia-dependent large EVs, named filopodia-derived vesicles (FDVs), can modify cellular behavior.
